1. A device for generating droplets, comprising:
- a body injection-molded as a single piece and including a bottom surface and a plurality of protrusions located opposite the bottom surface, each protrusion including an annular wall that tapers in thickness in a direction away from the bottom surface, a channel network being formed in the bottom surface and including a first channel, a second channel, and a third channel each arranged parallel to the bottom surface and meeting one another in a droplet-generation region;
a plurality of wells including a carrier well configured to supply carrier fluid to the droplet-generation region via the first channel, a sample well configured to supply sample-containing fluid to the droplet-generation region via the second channel, and a droplet well configured to receive sample-containing droplets in carrier fluid from the third channel, wherein each well of the plurality of wells has an upper portion created by one of the protrusions; and
a sealing member attached to the bottom surface of the body in a fluid-tight seal, and providing a bottom wall for each channel.

Abstract
A system, including method and apparatus, for generating droplets suitable for droplet-based assays. The disclosed systems may include either one-piece or multi-piece droplet generation components configured to form sample-containing droplets by merging aqueous, sample-containing fluid with a background emulsion fluid such as oil, to form an emulsion of sample-containing droplets suspended in the background fluid. In some cases, the disclosed systems may include channels or other suitable mechanisms configured to transport the sample-containing droplets to an outlet region, so that subsequent assay steps may be performed.

10. A device for generating droplets, comprising:

a substantially planar substrate having a top surface opposite a bottom surface; a plurality of protrusions injection-molded along with the substrate and projecting upwardly from the top surface of the substrate; an array of wells including a row of carrier wells configured to supply carrier fluid, a row of sample wells configured to supply sample-containing fluid, and a row of droplet wells configured to collect sample-containing droplets disposed in carrier fluid, wherein each well of the array of wells has an upper portion created by one of the protrusions, and wherein the carrier wells are larger than the sample wells and smaller than the droplet wells; a plurality of separate channel networks formed in the bottom surface of the substrate, each channel network interconnecting one of the carrier wells, one of the sample wells, and one of the droplet wells, and including a first channel, a second channel, and a third channel that meet one another in a droplet-generation region; and a sealing member attached to the bottom surface of the substrate in a fluid-tight seal, the sealing member providing a floor for each well and a bottom wall for each channel. - View Dependent Claims (11, 12, 13, 14, 15, 16, 17, 18, 19, 20, 21, 22, 23, 24)

25. A device for generating droplets, comprising:

a body injection-molded as a single piece and including a substrate having a bottom surface, the body also including a plurality of annular protrusions projecting from the substrate opposite the bottom surface, a channel network being formed in the bottom surface and including a first channel, a second channel, and a third channel each arranged parallel to the bottom surface; a plurality of wells including a carrier well configured to supply carrier fluid, a sample well configured to supply sample-containing fluid, and a droplet well configured to collect sample-containing droplets disposed in carrier fluid, wherein each well has an upper portion created by one of the protrusions; and a sealing member attached to the bottom surface of the substrate in a fluid-tight seal, the sealing member providing a bottom wall for each channel; wherein the first channel, the second channel, and the third channel meet one another in a droplet-generation region, and wherein the carrier well is fluidically connected to the droplet-generation region via the first channel, the sample well is fluidically connected to the droplet-generation region via the second channel, and the droplet well is fluidically connected to the droplet-generation region via the third channel.
