Abstract

The present disclosure relates to polymer coatings covalently attached to the surface of a substrate and the preparation of the polymer coatings, such as poly(N-(5-azidoacetamidylpentyl)acrylamide-co-acrylamide) (PAZAM), in the formation and manipulation of substrates, such as molecular arrays and flow cells. The present disclosure also relates to methods of preparing a substrate surface by using beads coated with a covalently attached polymer, such as PAZAM, and the method of determining a nucleotide sequence of a polynucleotide attached to a substrate surface described herein.

Claims

1. A substrate having a surface comprising a polymer coating covalently attached thereto, wherein the polymer coating comprises a polymer, said polymer comprising a recurring unit of Formula (Ia) and a recurring unit of Formula (II):

2. The substrate of claim 1, wherein R^A is selected from the group consisting of amine, optionally substituted alkene, optionally substituted alkyne, oxo-amine, azido, formyl, halo, hydroxyl, thiocyanate, carboxylic acid, epoxy, and thiol.

3. The substrate of claim 1, wherein R¹ is H or alkyl.

4. The substrate of claim 1, wherein R^A is azido.

5. The substrate of claim 1, wherein o is 5.

6. The substrate of claim 1, wherein:

R⁴ is —

C(O)NR⁷R⁸; and

R^4′

, R⁵, and R^5′

are each independently H or alkyl.

7. The substrate of claim 6, wherein R⁷ and R⁸ are each independently H or alkyl.

8. The substrate of claim 7, wherein R⁷ and R⁸ are each H.

9. The substrate of claim 1, wherein the functional groups on the surface comprise alkene or nitrene.

10. The substrate of claim 1, wherein the functional groups on the surface comprise activated ester and R^A is amine.

11. The substrate of claim 1, wherein the recurring unit of Formula (Ia) is

13. The substrate of claim 1, wherein the surface comprises both polymer-coated regions and inert regions.

14. The substrate of claim 13, wherein the inert regions are selected from the group consisting of glass regions, metal regions, mask regions, and interstitial regions.

15. The substrate of claim 14, wherein the inert regions comprise glass regions.

16. The substrate of claim 1, wherein the surface of the substrate comprises open wells.

17. The substrate of claim 1, wherein the substrate is a bead.

18. The substrate of claim 1, wherein the substrate is part of a flow cell.

19. The substrate of claim 3, wherein R^A is azido.

20. The substrate of claim 19, wherein:

, R⁵, and R^5′

are each independently H or alkyl.
