1. A device for the propagation or storage of microorganisms comprising:
- a) a first layer that comprises a first portion of a surface of the first layer, to which a first water-swellable gelling agent comprising a first clay is affixed, wherein the first clay comprises unmodified hectorite, organic-modified hectorite, or a combination thereof and the first layer is a water-proof substrate comprising a polymeric layer or paper with a water-proof coating; and
b) a second layer that is separable from the first layer and comprises a first portion of a surface of the second layer, to which a second water-swellable gelling agent is affixed.

Abstract
Devices for the propagation or storage of microorganisms are provided including a first layer that has a first portion of a surface of the first layer, to which a first water-swellable gelling agent comprising a first clay is affixed. The devices further include a second layer that is separable from the first layer and has a first portion of a surface of the second layer, to which a second water-swellable gelling agent is affixed. Methods for detecting and enumerating at least one microorganism in a sample are provided. The methods include providing a device, separating the first layer from the second layer, adding an aliquot of a sample containing at least one microorganism onto the first or second water-swellable gelling agent to form an inoculated device, laminating the first layer back to the second layer, and incubating the inoculated device. Kits and methods of making the devices are also provided.

13. A method of making a device for the propagation or storage of microorganisms comprising:

providing a first layer comprising a surface, wherein the first layer is a water-proof substrate comprising a polymeric layer or paper with a water-proof coating; affixing a first water-swellable gelling agent comprising a first clay to a first portion of the surface of the first layer, wherein the first clay comprises unmodified hectorite, organic-modified hectorite, or a combination thereof; providing a second layer comprising a surface; affixing a second water-swellable gelling agent to a first portion of the surface of the second layer; and laminating the first layer to the second layer, wherein the first water-swellable gelling agent at least partially contacts the second water-swellable gelling agent. - View Dependent Claims (14, 15, 16, 17)
