1. An assay system for conducting an assay on a fluid sample, the assay system comprising:

  • (a) a microfluidic cartridge comprising(i) a sandwich of three layers of substrate wherein a middle layer of substrate is disposed between and adhered to a top layer of substrate and a bottom layer of substrate, the three layers of substrate together defining one or more microfluidic channels disposed therein,(ii) a magnetic binding agent and a fluorescently labeled binding agent disposed within said channel(s), wherein (a) the magnetic binding agent comprises a first binding moiety coupled to a magnetic or paramagnetic particle, (b) the fluorescently labeled binding agent comprises a second, different binding moiety coupled to a fluorescent label, and (c) the first binding moiety and the second binding moiety bind an analyte to be detected so that, if analyte is present, the magnetic binding agent, the analyte, and the fluorescently labeled binding agent form a complex comprising the magnetic binding agent, the analyte and the fluorescently labeled binding agent,(iii) a sample inlet port for introducing said fluid sample into the cartridge,(iv) at least one port for allowing one or more fluids to be introduced into the cartridge from an associated reader device and transported through the microfluidic channel(s) and/or for allowing air to be vented from the cartridge, and(v) a fluid outlet sink for removing fluid from said channel(s), wherein, prior to use in the system, the cartridge does not contain a wash liquid; and

    (b) a reader device, the reader device comprising(i) a receiving port for introducing the cartridge into the reader,(ii) a pump configured to perform an air wash to move unbound label and/or sample from a detection area in said microfluidic channel,(iii) a magnet or system for generating a magnetic field such that, upon application of the magnetic field or moving the magnet of the reader into close proximity with the magnetic binding agents within said microfluidic channel the magnetic binding agent is held within the detection area in said microfluidic channel by the magnetic field or the magnet during the air wash and during detection in an air environment, and(iv) a detector configured to facilitate fluorescent detection of, in an air environment, analyte present within the one or more of the complex(es) held in the detection area by the magnetic field or the magnet.
