Abstract

The present disclosure provides a reagent cartridge configured for use in an automated multi-module cell processing environment.

Claims

1. An automated multi-module cell processing instrument comprising:

a housing;

a reagent cartridge, wherein the reagent cartridge comprises;

a thermal block with block reservoirs defined therein;

a cover disposed upon the thermal block with cover reservoirs defined therein, wherein the cover reservoirs and block reservoirs are coincident and together define reservoirs in the reagent cartridge;

an insert comprising a transformation module, wherein the transformation module is a flow-through electroporation device;

a script readable by a processor;

at least two additional inserts configured to reside within the reservoirs in the reagent cartridge; and

reagents dispensed in one or more of the reservoirs;

a growth module;

a cell concentration module;

an editing module; and

the processor.

2. The automated multi-module cell processing instrument of claim 1, wherein the one or more reservoirs with reagents dispensed therein is sealed with a foil seal.

3. The automated multi-module cell processing instrument of claim 1, further comprising a top cover disposed upon the cover wherein the top cover is configured to seal the reservoirs.

4. The automated multi-module cell processing instrument of claim 1, wherein the growth module comprises a rotating growth vial with media and cells dispensed therein.

5. The automated multi-module cell processing instrument of claim 4, wherein the cells are bacterial cells.

6. The automated multi-module cell processing instrument of claim 5, wherein the cells are E. coli cells.

7. The automated multi-module cell processing instrument of claim 4, wherein the cells are yeast cells.

8. The automated multi-module cell processing instrument of claim 7, wherein the cells are S. cerevisiae cells.

9. The automated multi-module cell processing instrument of claim 4, wherein the cells are plant cells.

10. The automated multi-module cell processing instrument of claim 4, wherein the cells are animal cells.

11. The automated multi-module cell processing instrument of claim 10, wherein the cells are mammalian cells.

12. The automated multi-module cell processing instrument of claim 1, wherein one of the reagents dispensed in the one or more reservoirs comprises an antibiotic.

13. The automated multi-module cell processing instrument of claim 1, wherein one of the reagents dispensed in the one or more reservoirs comprises a nucleic acid-guided nuclease.

14. The automated multi-module cell processing instrument of claim 13, wherein the nucleic acid-guided nuclease is MAD7.

15. The automated multi-module cell processing instrument of claim 1, wherein the editing module is configured to singulate cells.

16. An automated multi-module cell processing instrument comprising:

a housing;

a reagent cartridge for an automated multi-module cell processing instrument, wherein the reagent cartridge comprises;

a thermal block with block reservoirs defined therein;

a cover disposed upon the thermal block with cover reservoirs defined therein, wherein the cover reservoirs and block reservoirs are coincident and together define reservoirs in the reagent cartridge;

an insert comprising a transformation module;

a script readable by a processor;

at least two additional inserts configured to reside within the reservoirs in the reagent cartridge; and

reagents dispensed in one or more of the reservoirs;

a growth module;

a cell concentration module;

a singulation and editing module; and

the processor.

17. The automated multi-module cell processing instrument of claim 16, wherein the singulation and editing module is a solid wall device.

18. The automated multi-module cell processing instrument of claim 16, wherein the transformation module in the reagent cartridge is a flow-through electroporation device.

19. An automated multi-module cell processing instrument comprising:

a housing;

a reagent cartridge for an automated multi-module cell processing instrument, wherein the reagent cartridge comprises;

a thermal block with block reservoirs defined therein;

a cover disposed upon the thermal block with cover reservoirs defined therein, wherein the cover reservoirs and block reservoirs are coincident and together define reservoirs in the reagent cartridge;

an insert comprising a transformation module;

a script readable by a processor;

at least two additional inserts configured to reside within the reservoirs in the reagent cartridge; and

reagents dispensed in one or more of the reservoirs;

a growth module;

a cell concentration module; and

the processor.

20. The automated multi-module cell processing instrument of claim 19, wherein the transformation module in the reagent cartridge comprises a flow-through electroporation device.
