Abstract

Preparations for transferring efficiently oligonucleotides necessary in antisense therapy or the like into animal cells so as to be useful in treatment for various diseases, which comprises a collagen as an essential component are provided.

Claims

1. (Amended) A preparation for transferring a desired oligonucleotide into a target cell, which comprises a soluble type 1 collagen derived from dermis or a soluble type 1 collagen prepared by genetic engineering as an essential component, said preparation releasing a 5 to 30 mer oligonucleotide.

2. A preparation according to claim 1, wherein the target cell is animal cell.

3. (Amended) A preparation according to claim 2, wherein the target cell is an organ or a tissue which require to be treated for a subject disease, or its cell around.

4. A preparation according to any one of claims 1 to 3, wherein the collagen is atelocollagen.

5. A preparation according to any one of claims 1 to 4, wherein the molecular weight of the collagen is from about 300,000 to about 900,000.

6. A preparation according to any one of claims 1 to 5, wherein the preparation is a sustained-release formulation.

8. (Amended) A preparation according to claim 6, wherein the oligonucleotide is a DNA or a DNA derivative.

9. (Amended) A preparation according to claim 6, wherein the oligonucleotide is an RNA or an RNA derivative.

10. (Amended) A preparation according to claim 1, wherein the oligonucleotide has at least one phosphorothioate bond.

11. (Added) A preparation according to claim 1, wherein the oligonucleotide contains as at least portion a base sequence that binds complementarily under physiological conditions to a sense or antisense strand of a gene encoding a protein that exhibits a physiological effect to disrupt the homeostasis of a living body.
