Abstract

Novel vaccines for use against β-hemolytic Streptococcus colonization or infection are disclosed. The vaccines contain an immunogenic amount of a variant of streptococcal C5a peptidase (SCP). Also disclosed is a method of protecting a susceptible mammal against β-hemolytic Streptococcus colonization or infection by administering such a vaccine. Enzymatically inactive SCP, and polynucleotides encoding these SCP proteins are further disclosed.

Claims

1. A composition comprising an enzymatically inactive streptococcal C5a peptidase (SCP) conjugated or linked to one or more peptides.

2. The composition of claim 1, wherein the peptide is keyhole limpet hemocyanin (KLH), bovine serum albumin (BSA), ovalbumin, human serum albumin, human gamma globulin, chicken immunoglobulin G or bovine gamma globulin.

3. The composition of claim 1, wherein the peptide is a protein of another pathogen.

4. The composition of claim 3, wherein the protein of another pathogen is a Streptococcus pnuemoniae protein.

5. The composition of claim 1, wherein the SCP comprises the specificity crevice or catalytic domain of SCP.

6. The composition of claim 5, wherein the SCP comprises the catalytic domain of SCP.

7. The composition of claim 6, wherein the SCP further comprises the region of SCP between the catalytic domain and the cell wall insert.

8. The composition of claim 5, wherein the SCP comprises the specificity crevice.

9. The composition of claim 1, wherein the SCP does not contain an SCP signal sequence.

10. The composition of claim 1, wherein the SCP does not contain an SCP signal sequence and does not contain an SCP cell wall insert.

11. The composition of claim 1, wherein the SCP has a modification at one or more of amino acid residues 260, 261, 262, 415, 416, 417, 130, 193, 295 or 512.

12. A composition comprising an enzymatically inactive streptococcal C5a peptidase (SCP) conjugated or linked to one or more polysaccharides.

13. The composition of claim 12, wherein the polysaccharide is a group A Streptococci polysaccharide, a group B Streptococci polysaccharide, a group C Streptococci polysaccharide or a group G Streptococci polysacharide.

14. The composition of claim 12, wherein the polysaccharide is a polysaccharide of another pathogen.

15. The composition of claim 14, wherein the polysaccharide of another pathogen is a Streptococcus pnuemoniae polysaccharide.

16. The composition of claim 12, wherein the SCP comprises the specificity crevice or catalytic domain of SCP.

17. The composition of claim 16, wherein the SCP comprises the catalytic domain of SCP.

18. The composition of claim 17, wherein the SCP further comprises the region of SCP between the catalytic domain and the cell wall insert.

19. The composition of claim 17, wherein the SCP comprises the specificity crevice.

20. The composition of claim 12, wherein the SCP does not contain an SCP signal sequence.

21. The composition of claim 12, wherein the SCP does not contain an SCP cell wall insert.

22. The composition of claim 12, wherein the SCP has a modification at one or more of amino acid residues 260, 261, 262, 415, 416, 417, 130, 193, 295 or 512.
