Abstract

The present invention relates to methods and compositions comprising compounds that treat pathophysiological conditions arising from inflammatory responses. In particular, the present invention is directed to compounds that inhibit or block glycated protein produced induction of the signaling-associated inflammatory response in endothelial cells. The present invention relates to compounds that inhibit smooth muscle proliferation. In particular, the present invention is directed to compounds that inhibit smooth muscle cell proliferation by modulating HSPGs such as Perlecan. The present invention further relates to the use of compounds to treat vascular occlusive conditions characterized by smooth muscle proliferation such as restenosis and atherosclerosis.

Claims

1. A medical device comprising:

a drug delivering or eluting member; and

a compound disposed on or within the drug delivering or eluting member, the compound having the formula Id;

wherein

G is selected from NH or O;

Z is selected from H or

wherein X¹ is selected from F or Cl, and X² is selected from OCH₃, NH₂, OC(O)CH₃, or OH;

A is selected from NR¹ or O;

20. The medical device as claimed in claim 1, wherein the drug delivering or eluting member is a stent.

The medical device as claimed in claim 1, wherein the drug delivering or eluting member is selected from a shunt, a colostomy bag attachment device, an ear drainage tube, a lead for a pace maker, a lead for an implantable defibrillator, a suture, a staple, an anastornosis device, a vertebral disk, a bone pin, a suture anchor, a hemostatic barrier, a clamp, a screw, a plate, a clip, a vascular implant, a tissue adhesive, a tissue sealant, a tissue scaffold, a bone substitute, an intraluminal device, a stent, or a vascular support.

5. The medical device as claimed in claim 9, wherein the composition further comprises:

a pharmaceutically acceptable carrier;

optionally, a pharmaceutically acceptable auxiliary;

optionally, a pharmaceutically acceptable preservative; and

optionally, a pharmaceutically acceptable excipient.

The medical device as claimed in claim 9, wherein the composition further comprises an agent selected from a chemotherapeutic agent, an immunosuppressive agent, a cytokine, a cytotoxic agent, a nucleolytic compound, a radioactive isotope, a receptor, a pro-drug activating enzyme, an anti-inflammatory agent, an antirheumatic agent, a cardiovascular agent, a toxin, or any combination thereof.

7. The medical device as claimed in claim 9, wherein the composition is in the form of a capsule, a powder, a granule, a solution, a suspension, an emulsion, a cream, a gel, a paste, a foam, a spray, an aerosol, a microcapsule, a liposome, a paste, or a mouthwash.

10. The medical device as claimed in claim 9, wherein the drug delivering or eluting member is a stent.

The medical device as claimed in claim 9, wherein the drug delivering or eluting member is selected from a shunt, a colostomy bag attachment device, an ear drainage tube, a lead for a pace maker, a lead for an implantable defibrillator, a suture, a staple, an anastornosis device, a vertebral disk, a bone pin, a suture anchor, a hemostatic barrier, a clamp, a screw, a plate, a clip, a vascular implant, a tissue adhesive, a tissue sealant, a tissue scaffold, a bone substitute, an intraluminal device, a stent, a tablet, a cachet, a bolus, a lozenge, a suppository, a pessary, a tamport, a transdermal patch, a pastille, or a vascular support.
