Abstract

The present invention provides polypeptides and polynucleotides involved in C1 assimilation in Methylophilus methylotrophus and methods of producing amino acids in microorganisms having enhanced or attenuated expression of these polypeptides and/or polynucleotides.

Claims

1. An isolated polynucleotide, which encodes a protein comprising the amino acid sequence of SEQ ID NO:

2. A vector comprising said isolated polynucleotide of claim 1.

3. A host cell comprising said isolated polynucleotide of claim 1.

4. The host cell of claim 3, which is a Methylophilus bacterium.

5. The host cell of claim 4, which is a Methylophilus methylotrophus bacterium.

9. A method of producing at least one amino acid comprising culturing the host cell of claim 3 for a time and under conditions suitable for producing the amino acid;

and

collecting the amino acid produced.

10. The method of claim 9, wherein said at least one amino acid is an L-amino acid.

11. The method of claim 9, wherein said host cell is a Methylophilus bacterium.

12. The method of claim 9, wherein said host cell is a Methylophilus methylotrophus bacterium.

13. An isolated polynucleotide comprising a nucleotide sequence of SEQ D NO:

17. The host cell of claim 16, which is a Methylophilus methylotrophus bacterium.

21. A method of producing at least one amino acid comprising

culturing the host cell of claim 15 for a time and under conditions suitable for producing the amino acid;

and

collecting the amino acid produced.

22. The method of claim 21, wherein said at least one amino acid is an L-amino acid.

23. The method of claim 21, wherein said host cell is a Methylophilus bacterium.

24. The method of claim 21, wherein said host cell is a Methylophilus methylotrophus bacterium.

25. An isolated polynucleotide, which hybridizes under stringent conditions to said isolated polynucleotide of claim 13.

26. A vector comprising the isolated polynucleotide of claim 25.

27. A host cell comprising the isolated polynucleotide of claim 25.

29. A method of producing at least one amino acid comprising

culturing the host cell of claim 27 for a time and under conditions suitable for producing the amino acid;

and

collecting the amino acid produced.

30. The method of claim 29, wherein said at least one amino acid is an L-amino acid.

31. The method of claim 29, wherein said host cell is a Methylophilus bacterium.

32. The method of claim 29, wherein said host cell is a Methylophilus methylotrophus bacterium.

33. An isolated polynucleotide, which is at least 95% identical to the polynucleotide of claim 13.

34. A vector comprising the isolated polynucleotide of claim 33.

35. A host cell comprising the isolated polynucleotide of claim 33.

37. A method of producing at least one amino acid comprising

culturing the host cell of claim 35 for a time and under conditions suitable for producing the amino acid;

and

collecting the amino acid produced.

38. The method of claim 37, wherein said at least one amino acid is an L-amino acid.

39. The method of claim 37, wherein said host cell is a Methylophilus bacterium.

40. The method of claim 37, wherein said host cell is a Methylophilus methylotrophus bacterium.
