- (a) conducting rolling circle amplification of a nucleic acid to produce an amplicon comprising about two to about one hundred linked complements of a nucleic acid;
wherein said amplicon is anchored to a substrate, such that said amplicon is individually optically resolvable; and
(b) determining a sequence of at least a portion of said nucleic acid.
