Abstract

This invention provides compounds of conjugates of therapeutic or active agents with RAP or a RAP polypeptide, their pharmaceutical compositions and methods for using the such compounds and compositions in the diagnosis, prophylaxis, or treatment of diseases and conditions, including particularly diseases of the central nervous system or lysosomal storage diseases.

3. A method of delivering a therapeutic or diagnostic/investigational agent into the central nervous system in a subject in need thereof, said method comprising:

administering to said subject a compound comprising a Receptor Associated Protein (RAP) amino acid sequence having at least 80% homology to SEQ ID NO;

1, wherein the RAP amino acid sequence retains binding to a lipoprotein receptor-related protein (LRP) receptor and is conjugated to the therapeutic or diagnostic/investigational agent.

4. The method of claim 3 wherein

the agent is a therapeutic agent.

5. The method of claim 3, wherein the subject is suffering from a neurological or psychological condition or disease.

6. The method of claim 5, wherein the subject is suffering from Alzheimer'"'"'s disease (AD), Parkinson'"'"'s disease (PD), multiple sclerosis (MS), or amyotrophic lateral sclerosis (ALS), schizophrenia, epilepsy, a neurological infection, or a neurological inflammatory condition.

7. The method of claim 5, wherein the subject is suffering from a brain tumor or tumor metastases in the brain and the therapeutic agent is a chemotherapeutic agent.

The method of claim 4 wherein the therapeutic agent is selected from the group consisting of neurotransmitters, glial-derived neurotrophic factor (GDNF), Nerve Growth Factor, Brain-Derived Neurotrophic Factor, Neurotrophin-3, Neurotrophin-4/5, aFGF, bFGF, CNTF, Leukaemia Inhibitory Factor, Cardiotrophin-1, TGFb, BMPs, GDFs, Neurturin, Artemin, Persephin, EGF, TGFa, Neuregulins, IGF-1, IGF-2, ADNF, PDGFs, caspase inhibitors, antibodies, antimicrobial agents and lymphokines.

15. The method of claim 7 wherein the chemotherapeutic agent is adriamycin, cispiatin, 5-fluorouracil, camptothecin, or paclitaxel.

16. The method of claim 3 wherein the agent is a diagnostic/investigational agent and comprises a detectable label.

17. The method of claim 3 wherein the agent is conjugated to the RAP amino acid sequence via covalent bonding.

18. The method of claim 3 wherein the agent is conjugated to the RAP amino acid sequence via a peptide linker.
