Abstract

A method and apparatus for the manipulation of colloidal particles and biomolecules at the interface between an insulating electrode such as silicon oxide and an electrolyte solution. Light-controlled electrokinetic assembly of particles near surfaces relies on the combination of three functional elements: the AC electric field-induced assembly of planar aggregates; the patterning of the electrolyte/silicon oxide/silicon interface to exert spatial control over the assembly process; and the real-time control of the assembly process via external illumination. The present invention provides a set of fundamental operations enabling interactive control over the creation and placement of planar arrays of several types of particles and biomolecules and the manipulation of array shape and size. The present invention enables sample preparation and handling for diagnostic assays and biochemical analysis in an array format, and the functional integration of these operations. In addition, the present invention provides a procedure for the creation of material surfaces with desired properties and for the fabrication of surface-mounted optical components.

76. An array of oligonucleotides comprising oligonucleotides of differing sequences attached to particles arranged on a substrate in a planar configuration, said particles being encoded with a chemical or physical characteristic that allows identification of the oligonucleotide attached thereto, and wherein said particles are permanently anchored to said substrate.

78. The array of oligonucleotides according to claim 76 or 77, wherein the oligonucleotides of differing sequences are designed to hybridize with complementary oligonucleotides.

79. The array of oligonucleotides according to claim 76 or 77, wherein the oligonucleotides of differing sequences are single nucleic acid strands.

80. The array of oligonucleotides according to claim 79, wherein the target nucleic acid strands comprise DNA derived from a human being or complementary or homologous to DNA derived from a human being.

81. The array of oligonucleotides according to claim 79, wherein the nucleic acid strands include fragments of DNA or RNA.

82. The array of oligonucleotides according to claim 76 or 77, wherein the substrate comprises a semiconductor.

83. The array of oligonucleotides according to claim 76 or 77, wherein the substrate comprises an electrode.

84. The array of oligonucleotides according to claim 76 or 77, wherein the chemical or physical characteristic comprises a chemical tag.

85. The array of oligonucleotides according to claim 84, wherein the chemical tag is capable of being interrogated optically.

86. The array of oligonucleotides according to claim 76 or 77, wherein the particles reside on the upper surface of the substrate.

87. The array of oligonucleotides according to claim 76 or 77, wherein the oligonucleotides of differing sequences are exposed to a contiguous liquid phase.

88. The array of oligonucleotides according to claim 76 or 77, wherein the oligonucleotides of differing sequences comprise sequences capable of hybridizing to complementary nucleic acid strands contained within a contiguous liquid phase contacting the array.

89. The array of oligonucleotides according to claim 76 or 77, further comprising a multiplicity of subarrays of oligonucleotides, wherein the location of each subarray on said substrate in combination with the encoding indicates the types of oligonucleotides located therein.

77. An array of oligonucleotides comprising oligonucleotides of differing sequences, each of said differing oligonucleotides being attached to a particle and each of said particles being encoded with a chemical or physical characteristic that allows identification of the oligonucleotide attached thereto, wherein said particles are arranged on a substrate in a planar configuration.
