Abstract

The invention includes benzisoxazole piperazine compositions and methods of using them for modulating sleep.

Claims

1. A compound of Formula IV:

2. The compound of claim 1, wherein R₁, R₂, R₃, and R₄ are each H.

3. The compound of claim 1, wherein R₁, R₃, and R₄ are each H.

4. The compound of claim 1, wherein R₁, R₂, and R₄ are each H.

5. The compound of claim 1, wherein at least one of R₂ and R₃ is not H.

6. The compound of claim 1, wherein R₂ and R₃ are not H.

7. The compound of claim 1, wherein at least one of R₂ and R₃ is CH₃ or OCH₃.

8. The compound of claim 1, wherein R₂ or R₃ is CH₃.

9. The compound of claim 1, wherein R₃ is OCH₃.

10. The compound of claim 1, wherein t is 1 or 2.

11. The compound of claim 1, wherein R₅ and R₆ are each H.

12. The compound of claim 1, wherein R₅ and R₆ are each methyl.

13. The compound of claim 1, wherein R₅ and R₆ are each ethyl.

14. The compound of claim 1, wherein R₅ and R₆ together with the carbon to which they are attached are connected to form a spiro cyclopropyl ring.

15. The compound of claim 1, wherein Z is COOH.

16. The compound of claim 1, wherein Z is selected from CONHSO₂-alkyl and CONHSO₂-heteroalkyl.

17. The compound of claim 16, wherein Z is CONHSO₂CH₃, CONHSO₂CH(CH₃)₂, or

18. A composition comprising a compound of Formula IV:

21. A method of modulating sleep in a subject, comprising administering to a subject in need of thereof a therapeutically effective amount of a composition of claim 18.

22. The method of claim 21, wherein the subject is a human.

19. A compound having the formula of Compound 1:

20. A composition comprising the compound of claim 19 and at least one pharmaceutically acceptable excipient.
