Abstract

A system for the detection of compounds, including a target biological organism or component from a sample, using one or more reactant that will bind to the biological organism or compound forming a Raman active product, concentrating the Raman active product, and detecting the Raman active product using Raman light scattering.

Claims

1. A system for the detection of a target biological organism or component from a sample comprising

one or more reactants that will bind to the biological organism or compound forming a Raman active product;

means to concentrate the Raman active product; and

a detection sub-system which utilizes Raman light scattering to detect the Raman active product.

2. The system of claim 1, wherein the one or more reactants include an antibody to the target biological organism or compound.

3. The system of claim 1, wherein the one or more reactants include a bacteriophage.

4. The system of claim 1, wherein the one or more reactants include a bacteriophage protein.

5. The system of claim 1, wherein the Raman light scattering is resonance Raman scattering.

6. The system of claim 1, wherein one or more of the reactants that bind to the target biological organism or compound are attached to a solid surface.

7. The system of claim 6, wherein the solid surface is a microtiter plate.

8. The system of claim 6, wherein the solid surface is a microtiter well.

9. The system of claim 6, wherein the solid surface is a latex microparticle.

10. The system of claim 6, wherein the solid surface is a magnetic microparticle.

11. The system of claim 1, wherein the means to concentrate the Raman active product includes a series of decant and wash steps.

12. The system of claim 1, wherein the detection sub-system includes a laser.

13. The system of claim 8, wherein the laser is selected to have a wavelength that corresponds to an absorption wavelength of the Raman active product.

14. The system of claim 12, wherein the laser has output in the visible range.

15. The system of claim 14, wherein the laser has output at 532 nm.

16. The system of claim 12, wherein the laser has output in the ultra-violet range.

17. The system of claim 12, wherein the laser has output in the infra-red range.

18. The system of claim 1, wherein which the detection sub-system includes focusing optics.

19. The system of claim 1, wherein the detection sub-system includes a spectrometer.

20. The system of claim 1, wherein the detection sub-system includes a photodetector.

21. The system of claim 1, wherein the system includes one or more micro-fluidic channels.

22. The system of claim 21, wherein the micro-fluidic channels are part of an integrated circuit.

23. The system of claim 21, wherein the system includes an array of micro-fluidic wells.

25. The system of claim 1 further including enzymes in addition to the one or more reactants to convert the combination of one or more reactants and the biological organism or compound to a Raman active product.

26. The system of claim 1, wherein the biological organism or component is a bacterium.

27. The system of claim 26, wherein the bacterium is Listeria.

28. The system of claim 26, wherein the bacterium is E-coli.

29. The system of claim 26, wherein the bacterium is Salmonella.

30. The system of claim 26, wherein the bacterium is Vibrio.

31. The system of claim 26, wherein the bacterium is Camphelbacter.

32. The system of claim 26, wherein the bacterium is Staphylococcus.

33. The system of claim 1, wherein the biological organism or component is a virus.

34. The system of claim 33, wherein the virus is HIV.

35. The system of claim 33, wherein the virus is Hepatitis.

36. The system of claim 33, wherein the virus is Adenovirus.

37. The system of claim 33, wherein the virus is Rhino virus.

38. The system of claim 1, wherein the biological organism or component is a protein.

39. The system of claim 1, wherein the biological organism or component is a metabolite.

40. The system of claim 1, wherein the biological organism or component is a hormone.

41. The system of claim 1, wherein the biological organism or component is a metabolic intermediates.

24. The system of claim 24, wherein the micro-fluidic wells are part of an integrated circuit.

42. A system for the detection of Listeria from a sample comprising:

means for mixing the sample with a Listeria specific antibody bound to a solid surface, the Listeria specific antibody being designed to react with and become bound to any Listeria present in the sample;

means to incubate the mixed sample and antibody to increase concentration of bound Listeria;

means to wash away unwanted material; and

a detection sub-system which utilizes Raman light scattering to detect the concentration of bound Listeria.

43. The system of claim 42, wherein a bacteriophage is used instead of an antibody.

44. The system of claim 42, wherein a bacteriophage protein is used instead of an antibody.

45. The system of claim 42 in which the Raman light scattering is resonance Raman scattering.

46. The system of claim 42, wherein the solid surface is a microtiter plate.

47. The system of claim 42, wherein the solid surface is a microtiter well.

48. The system of claim 42, wherein the solid surface is a latex microparticle.

49. The system of claim 42, wherein the solid surface is a magnetic. microparticle.

50. The system of claim 42, wherein the detection sub-system includes a laser.

51. The system of claim 42, wherein the detection sub-system includes a laser adjusted to an absorption wavelength of the Raman active product.

52. The system of claim 50, wherein the laser has output in the visible range.

53. The system of claim 52, wherein the laser has output at 532 nm.

54. The system of claim 50, wherein the laser has output in the ultra-violet range.

55. The system of claim 50, wherein the laser has output in the infra-red range.

56. The system of claim 42, wherein the detection sub-system includes focusing optics.

57. The system of claim 42, wherein the detection sub-system includes a spectrometer.

58. The system of claim 42, wherein the detection sub-system includes a photodetector.

59. The system of claim 42, wherein the system includes one or more micro-fluidic channels.
