Abstract

Compositions for use in the treatment of wounds and fibrosis comprising a therapeutically effective amount of a compound that modulates actin assembly and organization intracellularly and/or extracellularly and a pharmaceutically acceptable vehicle; such a compound promotes the rate of wound healing and also prevents or reduces fibrosis. A preferred compound is gelsolin.

26. A method of reducing fibrosis in non-pulmonary tissue or wounds, the method comprising delivering directly to a tissue in need of such treatment a therapeutically effect amount of a compound that caps the fast growing end of actin filaments and/or severs capped actin filaments.

27. The method according to claim 26 wherein the compound is selected from the group consisting of gelsolin, villin, CapG, adseverin, flightless-1, advillin and derivatives thereof.

28. The method according to claim 27 wherein the compound is gelsolin or a derivative thereof.

29. The method according to claim 26 wherein scar formation is inhibited or prevented.

30. The method according to claim 29 wherein said tissue is eye tissue, nervous tissue or intestinal tissue.

31. The method according to claim 29 wherein dermal scarring is inhibited or prevented.

32. The method according to claim 29 wherein scar formation following a burn is inhibited or prevented.

33. The method according to claim 26, wherein said method is a method for reducing fibrosis in a wound such that the rate of healing of said wound is increased.

34. The method according to claim 33 wherein said wound is an acute wound.

35. The method according to claim 34 wherein said acute wound is a penetrative injury, a burn, nerve damage or a wound resulting from elective surgery.

36. The method according to claim 33 wherein said wound is a chronic wound.

37. The method according to claim 36 wherein said chronic wound is a diabetic, venous or decubitus ulcer or a wound of a generally healing compromised individual.

38. The method according to claim 26, wherein said method is a method of reducing fibrosis in non-pulmonary tissue.

39. The method according to claim 38 wherein the fibrosis is a fibrotic disorder.

40. The method according to claim 39 wherein the fibrotic disorder is selected from the group consisting of glomerulonephritis, cirrhosis of the liver and fibrocytic disease.

41. A delivery system for use in a gene therapy technique, said delivery system comprising a DNA molecule encoding a protein which modulates actin assembly and organization, said DNA molecule being capable of being transcribe to lead to the expression of said protein.

42. The delivery system according to claim 41 wherein the DNA molecule encodes one of gelsolin, villin, CapG, adseverin, flightless-1, advillin and derivatives thereof.

43. The use of a delivery system according to claim 41 for use in the manufacture of a medicament for use in the treatment of fibrosis in non-pulmonary tissue or wounds.

44. A method of treating fibrosis of non-pulmonary tissue or wounds comprising administering to a patient in need of treatment a therapeutically effective amount of a delivery system according to claim 43.
