Abstract
Methods, compositions and articles of manufacture involving cationic conjugated conformationally flexible polymers are provided. A method for the synthesis of cationic water-soluble polymers with linkages along the polymer main chain structure which disrupt the ability of the polymers to form extended-rod structures is provided. Such polymers may serve in the fabrication of novel optoelectronic devices and in the development of highly efficient biosensors. The invention further relates to the application of these polymers in assay methods.

25. An assay method comprising:
-
providing a sample that is suspected of containing a target molecule;
providing a signaling chromophore;
providing a conformationally flexible cationic conjugated polymer (CCP) that upon excitation is capable of transferring energy to the signaling chromophore, said CCP comprising an aromatic monomeric unit having bonds to its two adjacent polymeric units which form an angle of less than about 155°
with respect to one another;
providing a sensor molecule that can bind to the target molecule, wherein a greater amount of emitted light is produced from the signaling chromophore upon excitation of the CCP in the presence of target;
contacting the sample with the signaling chromophore, the sensor and the CCP in a solution under conditions in which the sensor can bind to the target, if present, and wherein the CCP can bind to the sensor and/or target, if present, wherein a signaling complex comprising the sensor, target, CCP and signaling chromophore is formed if the target is present;
applying a light source that can excite the CCP; and
detecting whether the light emitted from the signaling chromophore is increased in the presence of the sample. - View Dependent Claims (26, 27, 28, 29, 30, 31, 36, 37, 38, 39, 40)

32. A target sensing solution comprising:

a signaling chromophore; and
a conformationally flexible cationic conjugated polymer (CCP) that is capable of transferring energy to the signaling chromophore upon excitation when brought into proximity thereto, said CCP comprising an aromatic monomeric unit having bonds to its two adjacent polymeric units which form an angle of less than about 155°
with respect to one another, wherein a greater amount of energy can be produced from the signaling chromophore in the presence of the target being sensed when the CCP is excited. - View Dependent Claims (33, 34, 43, 44)

35. A kit for assaying a sample for a target biomolecule comprising:

a sensor biomolecule that can bind to the target biomolecule;
a signaling chromophore;
a conformationally flexible cationic conjugated polymer (CCP) that is capable of transferring energy to the signaling chromophore upon excitation when brought into proximity thereto, said CCP comprising an aromatic monomeric unit having bonds to its two adjacent polymeric units which form an angle of less than about 155°
with respect to one another, wherein said sensor interacts with the CCP and a detectibly greater amount of emitted light is produced from the signaling chromophore upon excitation of the CCP in the presence of target; and
a housing for retaining the reagents of the kit.
