Abstract

To provide a measuring probe excellent in sensitivity and reproducibility, simple operation in the protein immobilization onto a support can prevent peeling or damage. A layer containing a protein is adsorbed onto a support surface to fully encircle the long axis of the support, and the adsorbed layer is treated for cross-linking with a cross-linking agent in a concentration of 1 to 5 moles to 1 mole of the protein.

Claims

1. A measuring probe for detecting a specific analyte in a sample in contact with the sample, the probe comprising a support, a layer of a protein, wherein the protein layer has a circular form, and the protein in the protein layer has been cross-linked by using a solution containing a cross-linking agent in a concentration 1 to 5 times as much as that of the protein.

2. The measuring probe according to claim 1, wherein the protein layer is formed to have a portion fully encircling an axis of the support.

3. The measuring probe according to claim 1, wherein the cross-linking agent is a bifunctional cross-linking agent, and the functional group for the cross-linking is selected from the group consisting of aldehyde, carboxylic acid and carboxylic acid ester.

4. The measuring probe according to claim 3, where the cross-linking agent is glutaraldehyde.

5. The measuring probe according to claim 1, wherein the protein contained in the protein layer is a component for detecting the analyte, the component being an enzyme or a receptor.

6. The measuring probe according to claim 1, wherein the protein contained in the protein layer is a component for detecting the analyte, the component being an antibody or an antigen or a fragment thereof.

7. The measuring probe according to claim 1, wherein a component for detecting the analyte is held on a subbing layer being the protein layer.

8. The measuring probe according to claim 7, wherein the detecting component is selected from the group consisting of antibody, antigen, enzyme, receptor, nucleobase, sugar chain and metal coordinate.

9. The measuring probe according to claim 1, wherein the support has a configuration selected from the group consisting of fiber-, brush- and mesh-like configurations.

10. A process of manufacturing a measuring probe that specifically detects a specific analyte in a sample in contact with the sample, the process comprising a step of forming on a surface of a support a layer having a circular form and containing a cross-linked protein, wherein the protein in the layer is cross-linked by using a solution containing a cross-linking agent in a concentration 1-5 times as much as that of the protein.

11. The process according to claim 10, wherein the protein layer is formed to have a portion fully encircling one axis of the support.

12. The process according to claim 10, wherein the protein contained in the protein layer is at least one selected from the group consisting of antibodies, antigens, enzymes and receptors, as a component for detecting the analyte.

13. The process according to claim 12, wherein the protein is an antibody or an antigen.

14. The process according to claim 10, wherein the layer is a subbing layer, and the process further comprises a step of immobilizing a detecting component on the subbing layer.

15. The process according to claim 10, wherein the layer contains a component selected from the group consisting of an antibody, an antigen, an enzyme, a receptor, a nucleobase, a sugar chain, and a metal coordinate for detecting the analyte.

16. A measuring probe for detecting presence or absence of a target substance in a sample or for determining a concentration thereof in contact with the sample, the probe comprising a support having a columnar optical waveguide, a protein layer formed on a surface of the optical waveguide, wherein the protein layer has a circular form, and the protein molecules in the protein layer have been cross-linked to each other with a cross-linking agent.

17. The measuring probe according to claim 16, wherein the protein layer contains at least one protein selected from the group consisting of an antibody, an antigen, an enzyme and a receptor protein as a component for detecting the target substance.

18. The measuring probe according to claim 16, wherein the protein layer serves as a subbing layer, and a component for detecting the target substance is held on the protein layer.

19. The measuring probe according to claim 16, wherein the component for detecting the target substance is selected from the group consisting of an antibody, an antigen, an enzyme and a receptor.

20. The measuring probe according to claim 16, wherein the detection of presence or absence of the target substance or determination of the concentration thereof is carried detecting a signal based on binding between the detection component and the target substance.

21. The measuring probe according to claim 16, wherein the cross-linking agent is a bifunctional cross-linking agent and the functional group for cross-linking is selected from the group consisting of aldehyde, carboxylic acid, and carboxylic acid ester.
