Abstract

Provided is a method for production of nanocrystalline hydroxyapatite particles, and nanocrystalline hydroxyapatite particles produced according to the method. The nanocrystalline hydroxyapatite particles exhibit substantially superior cell transformation abilities as compared to known and commercially-available calcium phosphate kits. The nanocrystalline hydroxyapatite particles also find use in tissue engineering applications, for example bone and tooth engineering and repair applications.

Claims

1. A method for making a hydroxyapatite complex, comprising reacting calcium ions with Na₃PO₄ in the presence of hydroxyl ions and a biomolecule, wherein the calcium ions and Na₃PO₄ combine to precipitate as hydroxyapatite.

2. The method of claim 1, wherein the biomolecule is a nucleic acid.

3. The method of claim 2, wherein the nucleic acid is selected from the group consisting of DNA, plasmid DNA (pDNA), RNA, antisense RNA, and interfering RNA.

4. The method of claim 3, wherein the DNA comprises a gene.

5. The method of claim 4, wherein the DNA comprises a bone morphogenetic protein gene.

6. The method of claim 3, wherein the DNA comprises a gene selected from the group consisting of rhBMP-2, VEGF, EGF, NGF, TGF-β

, FGF, PDGF, IGF, Runx2, Osx, and BMP-7.

7. The method of claim 1, further comprising associating the hydroxyapatite with a substrate.

8. The method of claim 1, wherein the hydroxyapatite is prepared according to the formula:

n(CaCl₂)+6(Na₃PO₄)+2NaOH→

Ca₁₀(PO₄)₆(OH)₂+20NaCl+(n−

10)CaCl₂,

wherein n is greater than 10.

9. The method of claim 8, wherein n is greater than about 100.

10. The method of claim 8, wherein the Ca₁₀(PO₄)₆(OH)₂ is precipitated by mixing a solution of calcium chloride with a solution of NaCl, KCl, Na₃PO₄, a sugar, and a buffer adjusted to pH 7.5 with NaOH.

11. The method of claim 8, wherein the Ca₁₀(PO₄)₆(OH)₂ is precipitated by mixing a solution of calcium chloride with a solution of about 280 mM NaCl, about 10 mM KCl, about 1.5 mM Na₃PO₄, about 12 mM dextrose, and about 50 mM HEPES adjusted to pH 7.5 with NaOH.

12. The method of claim 1, wherein the hydroxyapatite has a size of 20 to 50 nanometers.

13. The method of claim 1, wherein the hydroxyapatite has a ratio of calcium to phosphate (Ca/P) of 100-300.

14. The method of claim 2, wherein the nucleic acid is condensed by greater than 70% when compared to the nucleic acid when it is not complexed with hydroxyapatite.

15. The method of claim 2, wherein the nucleic acid is bound with greater than 90% efficiency.

16. A composition, comprising a complex of hydroxyapatite and a biomolecule wherein the hydroxyapatite has a size of 20 to 50 nanometers.

17. The composition of claim 16, wherein the biomolecule is selected from the group consisting of a nucleic acid, a protein, and any combination thereof.

18. A composition comprising the complex of claim 17 and a polymer matrix.

19. The composition of claim 18, wherein the polymer matrix is selected from the group consisting of fibrin, collagen, glycosaminoglycans, chitin, chitosan, hyaluronic acid, polysaccharides, starch, carrageenan, alginate, heparin, glycogen, cellulose, polylactide (PLA), polylactide-co-glycolide (PLGA), polyglycolic acid (PGA), polyurethane, polycaprolactone, polymethyl methacrylate (PMMA), polyamino acids, poly-L-lysine, polyethyleneimine, poly-anhydrides, polypropylene-fumarate, polycarbonates, polyamides, polyanhydrides, polyortho esters, polyacetals, polycyanoacrylates, degradable polyurethanes, and combinations of any thereof.

20. The composition of claim 16, wherein the hydroxyapatite has a ratio of calcium to phosphate (Ca/P) of 100-300.

21. The composition of claim 20, wherein the hydroxyapatite is prepared according to the formula:

n(CaCl₂)+6(Na₃PO₄)+2NaOH→

Ca₁₀(PO₄)₆(OH)₂+20NaCl+(n−

10)CaCl₂,

wherein n is greater than 10.

22. The composition of claim 21, wherein n is greater than about 100.

23. A method for introducing a biomolecule into a cell, comprising contacting a cell with a composition comprising a complex of hydroxyapatite and a biomolecule wherein the complex has a size of 20 to 50 nanometers.
