Abstract

The present invention is directed to composite devices comprising a tissue-engineering scaffold and an anionic polysaccharide/structural protein coating. The composite devices are useful for the reparation and regeneration of tissue, particularly in the treatment of chronic wounds.

Claims

1. A composite device for the treatment of chronic wounds comprising, a tissue-engineering scaffold and an anionic polysaccharide/structural protein coating, wherein said tissue-engineering scaffold is a biocompatible, biodegradable polymer and said anionic polysaccharide is a mucopolysaccharide or polycarboxylate.

2. The composite device of claim 1, wherein said scaffold is selected from the group consisting of sponges, foams, perforated films and textiles.

3. The composite device of claim 2, wherein said scaffold is a textile selected from the group consisting of weaves, knits, braids, meshes and nonwovens.

4. The composite device of claim 3, wherein said scaffold is a nonwoven comprising a copolymer of glycolide and lactide.

5. The composite device of claim 3, wherein said scaffold is a nonwoven comprising a copolymer of glycolide and lactide having a mole ratio of glycolide to lactide of about 90:

6. The composite device of claim 2, wherein said scaffold is a foam comprising an elastomeric copolymer.

7. The composite device of claim 6, wherein said elastomeric copolymer comprises epsilon-caprolactone and glycolide.

8. The composite device of claim 7, wherein said elastomeric copolymer comprises a mole ratio of epsilon-caprolactone to glycolide of from about 30:

9. The composite device of claim 1, wherein said anionic polysaccharide is selected from the group consisting of heparin, hyaluronic acid, heparan sulphate, chondroitin sulphate, alginates, oxidized regenerated cellulose and oxidized cellulose.

10. The composite device of claim 1, wherein said anionic polysaccharide is selected from the group consisting of oxidized regenerated cellulose, oxidized cellulose, hyaluronic acid and chondroitin sulphate.

11. The composite device of claim 1, wherein said anionic polysaccharide is oxidized regenerated cellulose.

12. The composite device of claim 1, wherein said structural protein is selected from the group consisting of collagen, fibrin, fibronectin and laminin.

13. The composite device of claim 1, wherein said structural protein is collagen.

14. The composite device of claim 1, wherein said anionic polysaccharide/structural protein has a mole ratio of anionic polysaccharide to structural protein from about 10:

15. The composite device of claim 1, wherein said scaffold is a foam comprising an elastomeric copolymer of epsilon-caprolactone and glycolide having a mole ratio of epsilon-caprolactone to glycolide from about 35:

19. A kit comprising, a sterile composite device of claim 1.

20. The composite device of claim 1, wherein said device is capable of increasing the presence of activated macrophages in said wound.

18. A method of treating a wound comprising,

a) contacting a wound with one or more composite devices, wherein said device(s) comprise a biocompatible, biodegradable tissue-engineering scaffold having an anionic polysaccharide/structural protein coating,

b) allowing said devices and said wound to remain in contact with each other, and

c) covering or closing said wound,

wherein said wound is treated.
