1. A method of site specifically labeling a protein comprising:

  • i) constructing a fusion protein comprising;

    (a) a protein comprising at least a single unprotected and uniquely reactive amino acid, and(b) at least one domain comprising one or more uniquely protected reactive amino acids,ii) reacting said unprotected amino acid of protein (a) with a first modifying agent so that a covalent linkage between said unprotected amino acid and said first modifying agent is formed, andiii) deprotecting said one or more reactive amino acids of domain (b) and reacting said one or more deprotected reactive amino acids with a second modifying agent so that a covalent linkage between said one or more deprotected reactive amino acids and said second modifying agent is formed.
