Abstract

Novel products comprising conditioned cell culture medium compositions and methods of use are described. The conditioned cell medium compositions of the invention may be comprised of any known defined or undefined medium and may be conditioned using any eukaryotic cell type. The medium may be conditioned by stromal cells, parenchymal cells, mesenchymal stem cells, liver reserve cells, neural stem cells, pancreatic stem cells and/or embryonic stem cells. Additionally, the cells may be genetically modified. A three-dimensional tissue construct is preferred. Once the cell medium of the invention is conditioned, it may be used in any state. Physical embodiments of the conditioned medium include, but are not limited to, liquid or solid, frozen, lyophilized or dried into a powder. Additionally, the medium is formulated with a pharmaceutically acceptable carrier as a vehicle for internal administration, applied directly to a food item or product, formulated with a salve or ointment for topical applications, or, for example, made into or added to surgical glue to accelerate healing of sutures following invasive procedures. Also, the medium may be further processed to concentrate or reduce one or more factors or components contained within the medium.

Claims

1. A method of making a composition, comprising:

(a) forming a conditioned medium comprising culturing fibroblasts in a nutrient medium sufficient to meet the nutritional needs required to grow the cells in vitro wherein the conditioned medium contains amounts approaching physiological levels of at least vascular endothelial growth factors (VEGF) and/or keratinocyte growth factor (KGF);

(b) removing the conditioned medium from the cultured fibroblasts; and

(c) combining the conditioned medium with an acceptable carrier to form the composition.

2. The method of claim 1, wherein the conditioned medium obtained from said cultured fibroblasts contains VEGF and KGF.

3. The method of claim 1, wherein culturing comprises a continuous flow system.

4. The method of claim 1, wherein culturing comprises a bioreactor system.

5. The method of claim 1, wherein the cultured fibroblasts are suspended or floated in the nutrient medium.

6. The method of claim 1, further comprising (d) processing said conditioned medium, wherein processing is concentrating, filtering, purifying, or a combination thereof.

7. The method of claim 1, wherein approaching physiological levels is performed by concentrating said conditioned medium.

8. The method of claim 1, further comprising using the conditioned medium to treat hair loss.

9. The method of claim 1, further comprising using the conditioned medium to stimulate hair growth.

10. The method of claim 1, wherein said conditioned medium additionally contains minoxidil, antibiotics or a combination thereof.

11. The method of claim 1, wherein the conditioned medium obtained from said cultured cells is used to prevent or treat an inflammatory disease or condition.

12. The method of claim 1, further comprising using the conditioned medium to prevent or treat an inflammatory disease or condition.

13. The method of claim 1, further comprising using the conditioned medium to prevent or treat a cosmetic defect.

14. The method of claim 13, wherein said cosmetic defect is a congenital defect or an acquired defect.

15. The method of claim 14, wherein said acquired defect is a medical condition that occurs post-trauma, post-surgery, or post-infection.

16. The method of claim 14, wherein said acquired defect is a post-medical procedure defect.

17. The method of claim 14, wherein said acquired defect is depressed scars, subcutaneous atrophy, keratotic lesions, enopthalmos in an unucleated eye, acne pitting of the face, linear scleroderma with subcutaneous atrophy, saddle-nose deformity, Romberg'"'"'s disease or unilateral vocal cord paralysis.

18. The method of claim 13, wherein said cosmetic defect is glabellar frown lines, deep nasolabial creases, circum-oral geographical wrinkles, sunken cheeks or mammary hypoplasia.

19. The method of claim 1, wherein said conditioned medium contains at least 3.2 ng/ml VEGF.

20. The method of claim 1, wherein said conditioned medium contains at least 1.67 ng/ml KGF.

21. The method of claim 1, wherein said conditioned medium further comprises at least 2.3 ng/ml G-CSF.

22. The method of claim 1, wherein said conditioned medium further comprises at least 0.9 ng/ml IL-8.

23. The method of claim 1, wherein said conditioned medium further comprises at least 0.8 ng/ml TGF-β

.

24. A conditioned medium comprising VEGF and KGF, wherein the medium is produced by culturing fibroblasts in a nutrient medium sufficient to meet the nutritional needs required to grow the cells in vitro wherein the conditioned medium contains VEGF and/or KGF at amounts approaching physiological levels so that a conditioned medium is formed, and removing the conditioned medium from the cultured cells.

25. The conditioned medium of claim 24, wherein the conditioned medium obtained from said cultured fibroblasts contains VEGF and KGF.

26. The conditioned medium of claim 24, further wherein said conditioned medium is processed, wherein processing is concentrating, filtering, purifying, or a combination thereof.

27. The conditioned medium of claim 24, wherein approaching physiological levels is performed by concentrating said conditioned medium.

28. The conditioned medium of claim 24, wherein the VEGF and KGF are present in concentrations effective to stimulate hair growth.

29. The conditioned medium of claim 24, wherein the VEGF and KGF are present in concentrations effective to treat hair loss.

30. The conditioned medium of claim 24, wherein the VEGF and KGF are present in concentrations effective to prevent or treat an inflammatory disease or condition.

31. The conditioned medium of claim 24, wherein the VEGF and KGF are present in concentrations effective to prevent or treat a cosmetic defect.

32. The conditioned medium of claim 24, further containing at least one additional cellular factor selected from the group consisting of fibroblast growth factors, epidermal growth factors, granulocyte/macrophage colony stimulating factors, bone morphogenetic proteins, transforming growth factors, human growth hormone, platelet derived growth factors, basic fibroblast growth factors, nerve growth factors, collagens, fibronectin, terascin, glycosaminologycans, decorin, secreted human dermal matrix proteins, essential amino acids and cytokines.

33. The conditioned medium of claim 24, wherein the conditioned medium contains at least 3.2 ng/ml VEGF.

34. The conditioned medium of claim 24, wherein the conditioned medium contains at least 1.67 ng/ml KGF.

35. A composition comprising the conditioned medium of claim 24 and an acceptable carrier.

36. The composition of claim 35, wherein said composition is an injectable composition or a topical composition.

37. The composition of claim 36, wherein said topical composition is an ointment, a cream, a hydrogel or a lotion.

38. A method of treating hair loss by administering to a subject a composition of claim 35.

39. A method of stimulating hair growth by administering to a subject a composition of claim 35.

40. A method of preventing or treating a cosmetic defect by administering to a subject a composition of claim 35.

41. The method of claim 40, wherein said cosmetic defect is a congenital defect or an acquired defect.

42. The method of claim 41, wherein said acquired defect is a medical condition that occurs post-trauma, post-surgery or post-infection.

43. The method of claim 41, wherein said acquired defect is a post-medical procedure defect.

44. The method of claim 41, wherein said acquired defect is depressed scars, subcutaneous atrophy, keratotic lesions, enopthalmos in an unucleated eye, acne pitting of the face, linear scleroderma with subcutaneous atrophy, saddle-nose deformity, Romberg'"'"'s disease or unilateral vocal cord paralysis.

45. The method of claim 40, wherein said cosmetic defect is glabellar frown lines, deep nasolabial creases, circum-oral geographical wrinkles, sunken cheeks or mammary hypoplasia.

46. A cosmeceutical composition comprising:

cell culture media, conditioned by;

(a) culturing fibroblasts in a nutrient medium sufficient to meet the nutritional needs required to grow the cells in vitro wherein the conditioned medium contains amounts approaching physiological levels of at least vascular endothelial growth factors (VEGF) and/or keratinocyte growth factor (KGF), so that a conditioned medium is formed;

(b) removing the conditioned medium from the cultured cells; and

(c) combining the conditioned medium with an acceptable carrier to form the composition.

47. The cosmeceutical composition of claim 46, further wherein said conditioned medium is (d) processed, wherein processing is concentrating, filtering, purifying, or a combination thereof.

48. The cosmeceutical composition of claim 46, wherein approaching physiological levels is performed by concentrating said conditioned medium.

49. The cosmeceutical composition of claim 46, wherein the conditioned medium obtained from said cultured fibroblasts contains VEGF and KGF.

50. The cosmeceutical composition of claim 46, wherein conditioned medium obtained from said cultured cells is concentrated and used to prevent or treat a cosmetic defect.

51. The cosmeceutical composition of claim 46, wherein conditioned medium obtained from said cultured cells is purified and used to prevent or treat a cosmetic defect.

52. The cosmeceutical composition of claim 46, wherein said composition is an injectable composition or a topical composition.

53. The cosmeceutical composition of claim 52, wherein said topical composition is an ointment, a cream, a hydrogel or a lotion.

54. A method of preventing or treating a cosmetic defect by administering to a subject a cosmeceutical composition of claim 46.

55. The method of claim 54, wherein said cosmetic defect is a congenital defect or an acquired defect.

56. The method of claim 55, wherein said an acquired defect is a medical condition that occurs post-trauma, post-surgery or post-infection.

57. The method of claim 55, wherein said acquired defect is a post-medical procedure defect.

58. The method of claim 55, wherein said an acquired defect is depressed scars, subcutaneous atrophy, keratotic lesions, enopthalmos in an unucleated eye, acne pitting of the face, linear scleroderma with subcutaneous atrophy, saddle-nose deformity, Romberg'"'"'s disease or unilateral vocal cord paralysis.

59. The method of claim 54, wherein said cosmetic defect is glabellar frown lines, deep nasolabial creases, circum-oral geographical wrinkles, sunken cheeks or mammary hypoplasia.

60. The conditioned medium of claim 46, wherein the conditioned medium contains at least 3.2 ng/ml VEGF.

61. The conditioned medium of claim 46, wherein the conditioned medium contains at least 1.67 ng/ml KGF.

62. A method of making a composition, comprising:

(a) forming a conditioned medium comprising culturing fibroblasts in a nutrient medium sufficient to meet the nutritional needs required to grow the cells in vitro, wherein said cultured fibroblasts form an extracellular matrix and secrete a desired level of extracellular products so that a conditioned medium is formed;

(b) removing the conditioned medium from the cultured fibroblasts; and

(c) combining the conditioned medium with an acceptable carrier to form the composition.

63. The method of claim 62, wherein the conditioned medium obtained from said cultured fibroblasts contains VEGF, KGF, or both.

64. The method of claim 62, wherein culturing comprises a continuous flow system.

65. The method of claim 62, wherein culturing comprises a bioreactor system.

66. The method of claim 62, wherein the cultured fibroblasts are suspended or floated in the nutrient medium.

67. The method of claim 62, further comprising (d) processing said conditioned medium, wherein processing is concentrating, filtering, purifying, or a combination thereof.

68. The method of claim 62, wherein said conditioned medium additionally contains minoxidil, antibiotics or a combination thereof.

69. The method of claim 62, further comprising using the conditioned medium to treat hair loss.

70. The method of claim 62, further comprising using the conditioned medium to stimulate hair growth.

71. The method of claim 62, further comprising using the conditioned medium to prevent or treat a cosmetic defect.

72. The method of claim 71, wherein said cosmetic defect is a congenital defect or an acquired defect.

73. The method of claim 72, wherein said acquired defect is a medical condition that occurs post-trauma, post-surgery, post-infection or post-medical procedure.

74. The method of claim 72, wherein said acquired defect is a post-medical procedure defect.

75. The method of claim 72, wherein said acquired defect is depressed scars, subcutaneous atrophy, keratotic lesions, enopthalmos in an unucleated eye, acne pitting of the face, linear scleroderma with subcutaneous atrophy, saddle-nose deformity, Romberg'"'"'s disease or unilateral vocal cord paralysis.

76. The method of claim 71, wherein said cosmetic defect is glabellar frown lines, deep nasolabial creases, circum-oral geographical wrinkles, sunken cheeks or mammary hypoplasia.

77. The method of claim 62, wherein said conditioned medium contains at least 3.2 ng/ml VEGF.

78. The method of claim 62, wherein said conditioned medium contains at least 1.67 ng/ml KGF.
