Abstract

Methods and compositions are disclosed for determining feline proBNP or fragments thereof in a sample. In one method, feline proBNP or fragments thereof are determined by providing a feline sample, contacting the sample with at least one antibody that binds an epitope in the region from amino acids 1 to 20 of feline proBNP, and determining the presence of the feline proBNP or fragments thereof present in the sample. Antibodies that bind feline proBNP and kits comprising such antibodies are also disclosed.

Claims

1. A method of detecting feline proBNP or a fragment thereof in a feline sample comprising

(a) providing a feline sample;

(b) contacting the sample with at least one antibody that binds an epitope in the region from amino acids 1 to 20 of feline proBNP; and

(c) detecting the binding of the antibody to the epitope whereby the feline proBNP or the fragment thereof in the sample is detected.

2. The method of claim 1, wherein the epitope comprises at least 3 amino acids.

3. The method of claim 1, wherein the at least one antibody is polyclonal.

4. The method of claim 1, wherein the at least one antibody is monoclonal.

5. The method of claim 1, wherein the sample is a blood sample or a urine sample.

6. The method of claim 1, wherein detecting the binding of the antibody to the epitope comprises radioimmunoassay, immune binding assay, Western blot, immunohistochemistry, or enzyme immunoassay.

7. The method of claim 1, further comprising determining the concentration of the feline proBNP or fragment thereof in the sample.

8. The method of claim 7, further comprising comparing the concentration of the feline proBNP or fragment thereof in the sample with the concentration of feline proBNP or fragment thereof in a sample from a healthy feline.

9. The method of claim 8, further comprising diagnosing cardiac insufficiency in a feline from which the sample was obtained if the concentration of the feline proBNP or fragment thereof in the sample is higher than the concentration of feline proBNP or fragment thereof in the sample from the healthy feline.

10. The method of claim 1, wherein contacting the sample with the at least one antibody occurs on a solid phase.

11. The method of claim 10, wherein the at least one antibody is bound to a solid phase.

12. The method of claim 10, wherein the feline proBNP or fragment thereof is bound to a solid phase.

13. An isolated antibody that binds an epitope in the region from amino acids 1 to 20 of feline proBNP.

14. The antibody of claim 13, wherein the epitope comprises at least 3 amino acids in the region from amino acids 1 to 20 of feline proBNP.

15. A kit for detecting feline proBNP or a fragment thereof comprising an antibody that binds an epitope in the region from amino acids 1 to 20 of feline proBNP and a label.

16. The kit of claim 15, wherein the label is peroxidase, biotin, fluorescent dye, gold colloid, or a radionuclide.

17. The kit of claim 15, further comprising a peptide comprising amino acids 1 to 20 of feline proBNP.

18. The kit of claim 15, further comprising at least one further antibody.

19. The kit of claim 18, wherein the at least one further antibody is labeled.

20. The kit of claim 15, wherein the antibody is bound to a solid phase.
