Abstract

The present invention relates to the use of stem cell factor (SCF) for the treatment of ischemic injured tissue such as in cardiovascular disease. The method involves administration of a nucleic acid encoding SCF, wherein the nucleic acid is delivered to the site of the injury and is incorporated into cells which then express the SCF.

Claims

1. A method for treating a subject suffering from an ischemic injury, comprising administering to the subject a nucleic acid encoding stem cell factor (SCF) wherein the nucleic acid is delivered to the site of the injury where it is taken up by cells and SCF is expressed.

2. The method of claim 1, wherein the SCF is a secretable form of SCF.

3. The method of claim 2, wherein the SCF comprises amino acids 26-165 of FIG. 1.

4. The method of claim 1, wherein the SCF is a membrane bound form of SCF.

5. The method of claim 4, wherein the SCF comprises amino acids 25-245 of FIG. 1.

6. The method of claim 1, wherein the SCF comprises both a membrane bound form and a secretable form.

7. The method of claim 1, wherein the SCF is human SCF.

8. The method of claim 1, wherein the nucleic acid is a vector.

9. The method of claim 8, wherein said vector is a viral vector.

10. The method of claim 9, wherein said viral vector is packaged within an infectious viral particle.

11. The method of claim 9, wherein the viral vector is selected from the group consisting of adenovirus, retrovirus, herpes simplex virus, bovine papilloma virus, adeno-associated virus, lentiviral vector, vaccinia virus, and polyoma virus.

12. The method of claim 9, wherein the viral vector is an adenovirus vector or an adeno-associated virus vector.

13. The method of claim 1, wherein the injury is associated with the cardiovascular system.

14. The method of claim 1, wherein the injury is associated with a condition selected from the group consisting of myocardial infarction, ischemic stroke, peripheral vascular disease, and heart failure.

15. The method of claim 1, wherein the nucleic acid is delivered to the site of the injury using a catheter.

16. The method of claim 1, wherein delivery is to ischemic heart tissue.

17. The method of claim 1, wherein the nucleic acid is delivered to the coronary arterial circulation for delivery to ischemic heart tissue.

18. The method of claim 1, wherein the nucleic acid is delivered to ischemic heart tissue using a device implanted in a wall of the heart.

19. The method of claim 1, wherein expression of SCF at the site of the injury acts to recruit cells to the site to effect repair.

20. The method of claim 1, farther comprising the step of administering one or more cytokines or chemokines.

21. The method of claim 20, wherein said cytokines are selected from the group consisting of G-CSF and GM-CSF.

22. The method of claim 1, wherein the subject is not administered cells.

23. The method of claim 1, further comprising the step of administering cells.

24. The method of claim 23, wherein said cells are stem cells.

25. The method of claim 24 wherein said stem cells are generated ex vivo from tissue taken from the subject.
