Abstract

The present invention provides nucleoside cyclic phosphoramidates of structural formula (I) which are precursors to inhibitors of RNA-dependent RNA viral polymerase. These compounds are precursors to inhibitors of RNA-dependent RNA viral replication and are useful for the treatment of RNA-dependent RNA viral infection. They are particularly useful as pre-cursors to inhibitors of hepatitis C virus (HCV) NS5B polymerase, as precursors to inhibitors of HCV replication, and/or for the treatment of hepatitis C infection. The invention also describes pharmaceutical compositions containing such nucleoside cyclic phosphoramidates alone or in combination with other agents active against RNA-dependent RNA viral infection, in particular HCV infection. Also disclosed are methods of inhibiting RNA-dependent RNA polymerase, inhibiting RNA-dependent RNA viral replication, and/or treating RNA-dependent RNA viral infection with the nucleoside cyclic phosphoramidates of the present invention.

Claims

R⁶ is hydrogen, C_1-16 alkyl, C_2-20 alkenyl, (CH₂)_nC_3-6 cycloalkyl, phenyl, benzyl, or adamantyl;

wherein alkyl, alkenyl, cycloalkyl, and adamantyl are optionally substituted with one to three substituents independently selected from halogen, hydroxy, carboxy, C_1-4 alkoxy; and

wherein phenyl and benzyl are optionally substituted with one to three substituents independently selected from halogen, hydroxy, cyano, C_1-4 alkoxy, trifluoromethyl, and trifluoromethoxy; and

R⁹ is hydrogen, C_1-8 alkylcarbonyl, or C_1-8 alkyloxycarbonyl.

5. The compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R¹ is methyl or fluoromethyl, and R² is hydroxy.

6. The compound of claim 5, or a pharmaceutically acceptable salt thereof, wherein R¹ is methyl.

7. The compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R¹ is methyl or fluoromethyl, and R² is fluoro.

The compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R⁵ is hydrogen and R⁴ is selected from the group consisting of hydrogen, methyl, ethyl, n-propyl, isopropyl, isobutyl, 2-methyl-1-propyl, hydroxymethyl, fluoromethyl, mercaptomethyl, carboxymethyl, carbamoylmethyl, 1-hydroxyethyl, 2-carboxyethyl, 2-carbamoylethyl, 2-methylthioethyl, 4-amino-1-butyl, 3-amino-1-propyl, 3-guanidino-1-propyl, 1H-imidazol-4-ylmethyl, phenyl, benzyl, 4-hydroxybenzyl, and 1H-indol-3-ylmethyl.

The compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R⁶ is C_1-8 alkyl, cyclohexyl, cyclopentyl, cyclohexylmethyl, 2-cyclohexylethyl, or 3-cyclohexyl-n-propyl, each of which is optionally substituted with one to three substituents independently selected from fluorine, hydroxy, and C_1-6 alkoxy.

10. The compound of claim 9, or a pharmaceutically acceptable salt thereof, wherein R⁶ is ethyl, butyl, or heptyl.

11. The compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R⁴ is methyl, R⁵ is hydrogen, and R⁶ is ethyl, butyl, or heptyl.

and pharmaceutically acceptable salts thereof.

16. A pharmaceutical composition comprising a compound of claim 1, or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier.

17. A method for the treatment of hepatitis C virus infection in a mammal, which comprises administering a compound of claim 1, or a pharmaceutically acceptable salt thereof, to the mammal.
