Abstract

The invention relates generally to methods of treating spasticity, rigidity, or muscular hyperactivity conditions by introducing a portion of an expanded population of neural stem cells into an area of a recipient spinal cord.

Claims

1. A method of treating a spasticity or rigidity condition in a subject in need thereof comprising:

a) isolating neural stem cells of human origin;

b) expanding in vitro the isolated neural stem cells in a dispersed adherent culture to form an expanded population of neural stem cells;

c) concentrating the expanded population of neural stem cells to form concentrated neural stem cells;

d) introducing said concentrated neural stem cells into an area of central nervous system tissue of the subject with reduced GABA-producing or glycine-producing neurons; and

e) generating GABA-producing or glycine-producing neurons in said tissue.

2. The method of claim 1, wherein the spasticity or rigidity condition is associated with paralysis.

3. The method of claim 1, wherein the spasticity or rigidity condition comprises neuronal injury resulting from reduced blood flow to the tissue.

4. The method of claim 3, wherein the neuronal injury is from ischemic spinal cord injury, thoracic/abdominal aorta surgery, anterior spinal artery syndrome, chronic spinal spondylosis, or traumatic spinal cord injury.

5. The method of claim 1, wherein the spasticity or rigidity condition comprises neuronal injury that results in loss of GABA-producing or glycine-producing neurons in spinal cord tissue.

6. The method of claim 1, wherein the neural stem cells are multipotential.

7. The method of claim 1, wherein the neural stem cells are derived from human embryonic stem cells.

8. The method of claim 1, wherein the neural stem cells are isolated from a central nervous system tissue.

9. The method of claim 8, wherein the central nervous system tissue comprises fetal spinal cord tissue.

10. The method of claim 9, wherein the fetal spinal cord tissue is obtained from a post-mortem fetus having a gestational age of about 6.5 to about 20 weeks.

11. The method of claim 1, wherein the step of expanding in vitro said neural stem cells includes culturing the neural stem cells in the absence of serum.

12. The method of claim 1, wherein the step of expanding in vitro said neural stem cells includes exposing the neural stem cell to at least one mitogen.

13. The method of claim 12, wherein the mitogen is selected from the group consisting of bFGF, EGF, TGF-alpha, aFGF and combinations thereof.

14. The method of claim 1, wherein the neural stem cells are expanded to confluence.

15. The method of claim 1, wherein the introduced neural stem cells generate at least 1,000 GABA-producing neurons in said tissue.

16. The method of claim 1, wherein said concentrating step c) comprises concentrating the expanded population to a density of about 1,000 cells/μ

l to about 200,000 cells/μ

l.

17. The method of claim 1, wherein said concentrating step c) comprises concentrating the expanded population to a density of about 5,000 cells/μ

l to about 50,000 cells/μ

l.

18. The method of claim 1, wherein said introducing step d) comprises multiple injections of the concentrated neural stem cells.

19. The method of claim 1, wherein at least 1,000 cells from said concentrated neural stem cells are introduced to said area of central nervous system tissue of the subject with reduced GABA-producing or glycine-producing neurons.

20. The method of claim 1, wherein at least 20% of the concentrated neural stem cells differentiate into neurons in the spinal cord of an animal.
