Abstract

The present invention relates to novel polypeptides according to caroase 01-05 or any functional equivalents of any of them, suitable for use in a method for preparing a food products having increased whiteness, the use of the enzyme to increase whiteness of at least part of a food product, a process for preparing a food product wherein the enzyme is used and the food product obtained.

Claims

1. An isolated polypeptide having bleaching activity and one or more characteristics selected from the group consisting of:

a. An isolated polypeptide according to any one of SEQ ID NO;

08-12 or functional equivalents of any of them;

b. An isolated polypeptide obtainable by expressing a polynucleotide according to any one of SEQ ID NO;

01-07 or functional equivalents of any of them or a vector comprising said polynucleotides or functional equivalents of any of them in an appropriate host cell, e.g. Aspergillus niger; and

c. A polypeptide comprising at least one of SEQ ID NO;

13-17.

2. An isolated polypeptide according to claim 1 according to SEQ ID NO:

08-12 or functional equivalents of any of them, which functional equivalents are at least 55% homologous to SEQ ID NO;

8-12.

3. An isolated polypeptide according claim 1 according to SEQ ID NO:

08-12 or functional equivalents of any of them, which functional equivalents are at least 65% homologous to SEQ ID NO;

8-12.

4. An isolated polypeptide according to claim 1 obtainable from Marasmius scorodonius.

9. An isolated polynucleotide encoding a polypeptide according to claim 1.

21. A recombinant host cell expressing a polynucleotide according to claim 1.

22. Use of the polypeptide according to claim 1 increasing whiteness of at least part of a food product compared to a food product for which the polypeptide is not used.

23. Process for the production of a food product whereby an intermediate form of said food product comprises a pigment, which process comprises adding at least one polypeptide according to claim 1 in an amount that is effective in directly converting said pigment into a form which results in increasing the whiteness of at least part of the food product compared to the food product for which said polypeptide is not added during its production.

24. Process according to claim 23 wherein the food product is made from flour, preferably wheat flour.

25. Process according to claim 23 wherein the food product is a dairy product.

26. Process according to claim 23 wherein the pigment is a carotenoid.

27. Process according claim 23 wherein the enzyme is added as an enzyme preparation derived from a microorganism or produced is situ by a microorganism capable of producing said enzyme.

28. Process according to claim 27 wherein the enzyme is added as an enzyme preparation derived from or produced in situ by a bacterium, a fungus or a yeast.

29. Process according to claim 28 wherein the fungus belongs to the genus Marasmius, preferably Marasmius scorodonius.

30. Process according to claim 23, wherein additionally an oxidoreductase is added during production of the food product.

31. A food product obtainable by the process according to claim 23.

32. Use of a polypeptide according to claim 1 directly convert pigments into a form that results in an increased whiteness of at least part of a food product.

33. Use of a polypeptide according to claim 1 to detergents or in enzymatic stone bleach processes.

5. An isolated polynucleotide hybridisable to a polynucleotide according to SEQ ID NO:

01-07 or functional equivalents thereof.

6. An isolated polynucleotide according to claim 5 hybridisable under high stringency conditions to a polynucleotide according to SEQ ID NO:

01-07 or functional equivalents thereof.

7. An isolated polynucleotide according to claim 5 obtainable from a fungus, preferably a filamentous fungus.

8. An isolated polynucleotide according to claim 7 obtainable from Marasmius, preferably M. scorodonius.

13. A vector comprising a polynucleotide sequence according to claim 5.

14. A vector according to claim 13 wherein said polynucleotide sequence is operatively linked with regulatory sequences suitable for expression of said polynucleotide sequence in a suitable host cell.

15. A vector according to claim 14 wherein said suitable host cell is a filamentous fungus or a bacterium.

16. A method for manufacturing a polynucleotide or a vector according to claim 13 comprising the steps of culturing a host cell transformed with said polynucleotide or said vector and isolating said polynucleotide or said vector from said host cell.

17. An isolated polypeptide obtainable by expressing a polynucleotide or a vector according to claim 13 in an appropriate host cell.

19. A method for manufacturing a polypeptide comprising the steps of transforming a suitable host cell with an isolated polynucleotide or a vector according to claim 13, culturing said cell under conditions allowing expression of said polynucleotide and optionally purifying the encoded polypeptide from said cell or culture medium.

20. A recombinant host cell comprising a polynucleotide according to or a vector according to claim 13.

10. An isolated polynucleotide encoding at least one functional domain of a polypeptide according to SEQ ID NO:

08-12 or functional equivalents thereof.

11. An isolated polynucleotide comprising a nucleotide sequence according SEQ ID NO:

01-07 or functional equivalents thereof.

12. An isolated polynucleotide according to SEQ ID NO:

01-07.

18. Recombinant bleaching enzyme comprising a functional domain of a caroase 01-05 polypeptide.
