Abstract

The present invention relates to a biologically active functionalized electrospun matrix to permit immobilization and long-term delivery of biologically active agents. In particular the invention relates to a functionalized polymer matrix comprising a matrix polymer, a compatibilizing polymer and a biomolecule or other small functioning molecule. In certain aspects the electrospun polymer fibers comprise at least one biologically active molecule functionalized with low molecular weight heparin. Examples of active molecules that may be used with the multicomponent polymer of the invention include, for example, a drug, a biopolymer, for example a growth factor, a protein, a peptide, a nucleotide, a polysaccharide, a biological macromolecule or the like. The invention is further directed to the formation of functionalized crosslinked matrices, such as hydrogels, that include at least one functionalized compatibilizing polymer capable of assembly.

Claims

1. A crosslinked matrix comprising at least two compatibilizing polymer-LMWH conjugates, wherein the compatibilizing polymer comprises PEG, and at least one crosslinking compound, wherein each compatibilizing polymer-LMWH conjugate is capable of assembly and comprises at least one compatibilizing polymer covalently bonded to at least one LMWH molecule, wherein each of the compatibilizing polymer-LMWH conjugates are partially noncovalently crosslinked with the same crosslinking compound, and wherein the crosslinking compound is an incorporated molecule comprising a heparin-binding growth factor or a heparin-binding compatibilizing polymer-peptide conjugate.

8. The crosslinked matrix of claim 1 wherein the crosslinking compound comprises a heparin-binding compatibilizing polymer-peptide conjugate.

10. The crosslinked matrix of claim 8 wherein the heparin-binding compatibilizing polymer-peptide conjugate comprises PF4_ZIP, HIP, or ATIII.

21. The crosslinked matrix of claim 1 wherein the LMWH has an average molecular weight of about 10,000 daltons.

22. The crosslinked matrix of claim 1 wherein the compatibilizing polymer comprises four-arm star PEG.

23. The crosslinked matrix of claim 1 wherein the LMWH has an average molecular weight of about 3,000 daltons.

24. The crosslinked matrix of claim 1 wherein the incorporated molecule release is directly correlated to the erosion of the matrix.

25. The crosslinked matrix of claim 1 wherein the incorporated molecule release is directly correlated to cell-receptor interactions with the matrix.

26. The crosslinked matrix of claim 8 wherein the heparin-binding compatibilizing polymer-peptide conjugate is capable of assembly and comprises at least one compatibilizing polymer covalently bonded to at least two heparin-binding peptides.

29. The crosslinked matrix of claim 1 wherein the crosslinking compound comprises a heparin-binding growth factor.

A crosslinked matrix comprising at least three compatibilizing polymer-LMWH conjugates, wherein the compatibilizing polymer comprises PEG, at least one incorporated molecule comprising a heparin-binding growth factor and at least one heparin-binding compatibilizing polymer-peptide conjugate wherein each compatibilizing polymer-LMWH conjugate is capable of assembly and comprises at least one compatibilizing polymer covalently bonded to at least one LMWH molecule, wherein at least two of the compatibilizing polymer-LMWH conjugates are partially noncovalently crosslinked with the same incorporated molecule, wherein at least two of the compatibilizing polymer-LMWH conjugates are partially noncovalently crosslinked with the same heparin-binding compatibilizing polymer-peptide conjugate.

28. The crosslinked matrix of claims 29 and 27 wherein the growth factor comprises fibroblast growth factor, vascular endothelial growth factor, heparin-binding epidermal growth factor, transforming growth factor, acidic fibroblast growth factor or basic fibroblast growth factor.
