Abstract

Methods of inhibiting phosphatidylinositol 3-kinase delta isoform (PI3Kδ) activity, and methods of treating diseases, such as disorders of immunity and inflammation, in which PI3Kδ plays a role in leukocyte function are disclosed. Preferably, the methods employ active agents that selectively inhibit PI3Kδ, while not significantly inhibiting activity of other PI3K isoforms. Compounds are provided that inhibit PI3Kδ activity, including compounds that selectively inhibit PI3Kδ activity. Methods of using PI3Kδ inhibitory compounds to inhibit cancer cell growth or proliferation are also provided. Accordingly, the invention provides methods of using PI3Kδ inhibitory compounds to inhibit PI3Kδ-mediated processes in vitro and in vivo.

17. The compound of claim 16, wherein R¹² is alkylphenyl.

18. The compound of claim 17, wherein R¹⁰ is CH₃ and R¹¹ is H.

19. The compound of claim 18, wherein R^d is NH₂.

20. The compound of claim 16, wherein the compound is 2-(6-aminopurin-9-ylmethyl)-5-methyl-3-o-tolyl-3H-quinazolin-4-one.

23. A method to treat an arthritic disorder, which comprises administering to a subject in need thereof an effective amount of a compound of claim 16.

24. The method of claim 23, wherein the arthritic disorder is selected from rheumatoid arthritis, monoarticular arthritis, osteoarthritis, and gouty arthritis.

25. The method of claim 23, wherein the compound is 2-(6-aminopurin-9-ylmethyl)-5-methyl-3-o-tolyl-3H-quinazolin-4-one or a pharmaceutically acceptable salt thereof.

26. A method to treat a condition selected from the group consisting of asthma, allergic rhinitis, chronic obstructive pulmonary disease, diabetes, and multiple sclerosis;

which comprises administering to a subject in need thereof an effective amount of a compound of claim 16.

27. The method of claim 26, wherein the compound is 2-(6-aminopurin-9-ylmethyl)-5-methyl-3-o-tolyl-3H-quinazolin-4-one or a pharmaceutically acceptable salt thereof.

28. A method to treat leukemia, which comprises administering to a subject in need thereof an effective amount of a compound of claim 16.

29. The method of claim 28, wherein leukemia is chronic myeloid leukemia (CML) or lymphocytic leukemia.

30. The method of claim 28, wherein the compound is 2-(6-aminopurin-9-ylmethyl)-5-methyl-3-o-tolyl-3H-quinazolin-4-one or a pharmaceutically acceptable salt thereof.

22. A pharmaceutical composition comprising an effective amount of a compound of 2-(6-aminopurin-9-ylmethyl)-5-methyl-3-o-tolyl-3H-quinazolin-4-one or a pharmaceutically acceptable salt thereof;

and a pharmaceutically acceptable excipient.
