Abstract

Provided herein are methods of protecting all or a portion of the optic nerve fiber layer within the optic nerve/lamina cribrosa (ONLC) complex in an individual in need thereof, comprising administering to the individual a therapeutically effective amount of an agent that results in crosslinking of collagen in the indiviudal'"'"'s peripapillary sclera, thereby stiffening the individual'"'"'s peripapillary sclera and protecting all or a portion of the optic nerve fiber layer within the ONLC of the individual. In a particular embodiment, the invention is directed to methods of treating glaucoma in an individual in need thereof, comprising administering to the individual a therapeutically effective amount of an agent that results in crosslinking of collagen in the indiviudal'"'"'s peripapillary sclera, thereby stiffening the individual'"'"'s peripapillary sclera and treating the glaucoma in the individual. In yet another embodiment, the invention is directed to methods of treating glaucomatous optic neuropathy in an individual in need thereof, comprising administering to the individual a therapeutically effective amount of an agent that results in crosslinking of collagen in the indiviudal'"'"'s peripapillary sclera, thereby stiffening the individual'"'"'s peripapillary sclera and treating the glaucomatous optic neuropathy in the individual. The method can further comprise administering to the individual a therapeutically effective amount of an agent that results in crosslinking of collagen in the indiviudal'"'"'s lamina cribrosa (LC).

Claims

1. A method of treating glaucoma in an individual in need thereof, comprising administering to the individual a therapeutically effective amount of an agent that results in crosslinking of collagen in the indiviudal'"'"'s peripapillary sclera, thereby stiffening the individual'"'"'s peripapillary sclera and treating the glaucoma in the individual.

5. The method of claim 1 wherein the agent is selected from the group consisting of:

a photosensitizer crosslinking agent and light, a chemical fixative crosslinking agent, a glycation-induced crosslinking agent, a nitro alcohol based agent and a combination thereof.

6. The method of claim 5 wherein the photosensitizer crosslinking agent is riboflavin or a customized flash-linking agent and the light is UVA light.

9. The method of claim 5 wherein the photosensitizer crosslinking agent is riboflavin and the light is blue light.

10. The method of claim 9 wherein the riboflavin concentration is from about 0.05% to about 0.2% and the blue light is at a wavelength of from about 430 nm to about 475 nm.

11. The method of claim 5 wherein the chemical fixative crosslinking agent is glutaraldehyde.

12. The method of claim 5 wherein the glycation-induced crosslinking agent is an aldehyde sugar or a nitrite containing solution.

13. The method of claim 12 wherein the aldehyde sugar is selected from the group consisting of:

methylglyoxal, glyceraldehyde, ribose, and a combination thereof.

A method of treating glaucomatous optic neuropathy in an individual in need thereof, comprising administering to the individual a therapeutically effective amount of an agent that results in crosslinking of collagen in the indiviudal'"'"'s peripapillary sclera, thereby stiffening the individual'"'"'s peripapillary sclera and treating the glaucomatous optic neuropathy in the individual.

19. The method of claim 15 wherein the agent is selected from the group consisting of:

a photosensitizer crosslinking agent and light, a chemical fixative crosslinking agent, a glycation-induced crosslinking agent, a nitro alcohol based agent and a combination thereof.

20. The method of claim 19 wherein the photosensitizer crosslinking agent is riboflavin or a customized flash-linking agent and the light is UVA light.

23. The method of claim 19 wherein the photosensitizer crosslinking agent is riboflavin and the light is blue light.

24. The method of claim 23 wherein the riboflavin concentration is from about 0.05% to about 0.2% and the blue light is at a wavelength from about 430 nm to about 465 nm.

25. The method of claim 19 wherein the chemical fixative crosslinking agent is glutaraldehyde.

26. The method of claim 19 wherein the glycation-induced crosslinking agent is an aldehyde sugar or a nitrite containing solution.

27. The method of claim 26 wherein the aldehyde sugar is selected from the group consisting of:

methylglyoxal, glyceraldehyde, ribose and a combination thereof.

A method of protecting all or a portion of the optic nerve fiber layer within the optic nerve/lamina cribrosa complex in an individual in need thereof, comprising administering to the individual a therapeutically effective amount of an agent that results in crosslinking of collagen in the indiviudal'"'"'s peripapillary sclera, thereby stiffening the individual'"'"'s peripapillary sclera and protecting all or a portion of the optic nerve fiber layer within the optic nerve/lamina cribrosa in the individual.

33. The method of claim 29 wherein the agent is selected from the group consisting of:

a photosensitizer crosslinking agent and light, a chemical fixative crosslinking agent, a glycation-induced crosslinking agent, a nitro alcohol based agent and a combination thereof.

34. The method of claim 33 wherein the photosensitizer crosslinking agent is riboflavin or a customized flash-linking agent and the light is UVA light.

37. The method of claim 33 wherein the photosensitizer crosslinking agent is riboflavin and the light is blue light.

38. The method of claim 37 wherein the riboflavin concentration is from about 0.05% to about 0.2% and the blue light is at a wavelength of from about 430 nm to about 475 nm.

39. The method of claim 33 wherein the chemical fixative crosslinking agent is glutaraldehyde.

40. The method of claim 33 wherein the glycation-induced crosslinking agent is an aldehyde sugar or a nitrite containing solution.

41. The method of claim 40 wherein the aldehyde sugar is selected from the group consisting of:

methylglyoxal, glyceraldehyde, ribose, and a combination thereof.
