Abstract

The present invention discloses methods and compositions comprising a biocompatible gel that is used in tissue repair and regeneration. Once the gel is administered to a tissue site in need of repair or regeneration, the cells migrate from the gel into the tissue and repair or regenerate the tissue.

Claims

1. A method of delivering cells to tissue in need of repair comprising:

a) mixing cells to be delivered with gel;

b) injecting the cell/gel mixture to the site in need of repair; and

c) allowing the cells to migrate from the gel to the tissue in need of rep air.

2. The method according to claim 1, wherein the cells are autologous cells, embryonic/mesenchymal stem cells, or progenitor cells.

3. The method according to claim 1, wherein the gel is crosslinked.

4. The method according to claim 3, wherein transglutaminase, Ca++ dependent transglutaminase, Ca++ independent transglutaminase, laccase, or tyrosinase is used to crosslink the gel.

5. The method according to claim 1, wherein the gel is a hydrogel.

6. The method according to claim 5, wherein the hydrogel is transglutaminase reactive with fibrin, fibrinogen, collagen, albumen, involucrin, or gelatin.

7. The method according to claim 5, wherein the hydrogel further comprises agents that are reactive with transglutaminase.

8. The method according to claim 5, wherein the hydrogel comprises synthetic peptides that are similar in structure to gelatin.

9. The method according to claim 5, wherein the hydrogel contains peptides that contain glutamine amino acids and/or lysine amino acids.

10. The method according to claim 1, wherein the tissue in need of repair comprises collagen.

11. The method according to claim 1, wherein the tissue in need of repair is cartilage, bone, tendon/ligament, meniscus, spinal cord, skin, tendon/ligament, intervertebral disc, or retinal.

12. A method of repairing damaged tissue comprising:

a) mixing cells to be delivered with gel;

b) injecting the cell/gel mixture to the site in need of repair;

c) allowing the cells to migrate from the gel to the damaged tissue; and

d) allowing the migrated cells to repair the damaged tissue.

The method according to claim 12, wherein the cells are autologous cells, embryonic/mesenchymal stem cells, or progenitor cells.

13. The method according to claim 12, wherein the gel is crosslinked.

14. The method according to claim 13, wherein transglutaminase, Ca++ dependent transglutaminase, Ca++ independent transglutaminase, laccase, or tyrosinase is used to crosslink the gel.

15. The method according to claim 12, wherein the gel is a hydrogel.

16. The method according to claim 15, wherein the hydrogel is transglutaminase reactive with fibrin, fibrinogen, collagen, albumen, involucrin, or gelatin.

17. The method according to claim 15 , wherein the hydrogel further comprises agents that are reactive with transglutaminase.

18. The method according to claim 15, wherein the hydrogel comprises synthetic peptides that are similar in structure to gelatin.

19. The method according to claim 15, wherein the hydrogel contains peptides that contain glutamine amino acids and/or lysine amino acids.

20. The method according to claim 12, wherein the tissue in need of repair comprises collagen.

21. The method according to claim 12, wherein the tissue in need of repair is cartilage, bone, tendon/ligament, meniscus, spinal cord, skin, tendon/ligament, intervertebral disc, or retinal.

22. A composition for delivering cells to tissue in need of repair comprising gel and cells.

23. The composition according to claim 22, wherein the cells are autologous cells, embryonic/mesenchymal stem cells, or progenitor cells.

24. The composition according to claim 22, wherein the gel is crosslinked.

25. The composition according to claim 24, wherein transglutaminase, Ca++ dependent transglutaminase, Ca++ independent transglutaminase, laccase, or tyrosinase is used to crosslink the gel.

26. The composition according to claim 22, wherein the gel is a hydrogel.

27. The composition according to claim 26, wherein the hydrogel is transglutaminase reactive with fibrin, fibrinogen, collagen, albumen, involucrin, or gelatin.

28. The composition according to claim 26 , wherein the hydrogel further comprises agents that are reactive with transglutaminase.

29. The composition according to claim 26, wherein the hydrogel comprises synthetic peptides that are similar in structure to gelatin.

30. The composition according to claim 26, wherein the hydrogel contains peptides that contain glutamine amino acids and/or lysine amino acids.

31. The composition according to claim 22, wherein the tissue in need of repair comprises collagen.

32. The composition according to claim 22, wherein the tissue in need of repair is cartilage, bone, tendon/ligament, meniscus, spinal cord, skin, tendon/ligament, intervertebral disc, or retinal.

33. A gel for delivering cells to tissue in need of repair or regeneration comprising gelatin and a crosslinking agent.

34. The gel according to claim 33, wherein the gel comprises 4-20% gelatin.

35. The gel according to claim 33, wherein the gel comprises 5-10% gelatin.

36. The gel according to claim 33, wherein the gel may be in the form of an injectable gel, precast gel, beads, or micelles.

37. The gel according to claim 33, wherein the gel is a hydrogel.

38. The hydrogel according to claim 37, wherein transglutaminase, Ca++ dependent transglutaminase, Ca++ independent transglutaminase, laccase, or tyrosinase is used to crosslink the hydrogel.

39. The gel according to claim 33, wherein the hydrogel is transglutaminase reactive with fibrin, fibrinogen, collagen, albumen, involucrin, or gelatin.

40. The gel according to claim 33, wherein the gel further comprises agents that are reactive with transglutaminase.

41. The gel according to claim 33, wherein the gel further comprises synthetic peptides that are similar in structure to gelatin.

42. The gel according to claim 33, wherein the gel comprises peptides that contain glutamine amino acids and/or lysine amino acids.

43. The gel according to claim 33, wherein the tissue in need of repair comprises collagen.

44. The gel according to claim 373 wherein the tissue in need of repair is cartilage, bone, tendon/ligament, meniscus, spinal cord, skin, tendon/ligament, intervertebral disc, or retinal.
