Abstract

Disclosed are antibodies that specifically recognize Δ5-desaturase, methods of producing the antibodies, nucleotides and polypeptides for producing the antibodies, and methods of using the antibodies. The Δ5-desaturase-specific antibodies provide improved methods of detecting Δ5-desaturase in a sample.

Claims

1. An antibody that specifically binds Δ

5-desaturase derived from fungus or algae.

2. The antibody of claim 1 wherein the antibody specifically binds Δ

5-desaturase derived from Saprolegnia diclina or Mortierella alpina.

3. The antibody of claim 2 wherein the antibody specifically binds Δ

5-desaturase having the polypeptide sequence of SEQ ID NO;

4. The antibody of claim 1 wherein the antibody specifically binds full-length Δ

5-desaturase derived from fungus or algae.

5. The antibody of claim 1 wherein the antibody is polyclonal.

6. The antibody of claim 1 wherein the antibody is monoclonal.

7. The antibody of claim 1 wherein the antibody specifically binds a Δ

5-desaturase polypeptide fragment derived from fungus or algae.

8. The antibody of claim 7 wherein the antibody specifically binds a polypeptide fragment derived from Saprolegnia diclina or Mortierella alpina.

9. The antibody of claim 8 wherein the antibody specifically binds a polypeptide fragment comprising at least 8 contiguous amino acid residues of SEQ ID NO:

2 or conservative variations of SEQ ID NO;

10. The antibody of claim 9 wherein the antibody specifically binds a polypeptide fragment comprising SEQ ID NO:

2 or conservative variations of SEQ ID NO;

11. The antibody of claim 7 wherein the antibody is polyclonal.

13. An antibody that specifically binds a polypeptide fragment comprising at least 8 contiguous amino acid residues of SEQ ID NO:

2 or conservative variations of SEQ ID NO;

14. The antibody of claim 13 wherein the antibody binds a polypeptide fragment comprising SEQ ID NO:

2 or conservative variations of SEQ ID NO;

15. The antibody of claim 13 wherein the antibody is polyclonal.

16. The antibody of claim 13 wherein the antibody is monoclonal.

17. A method of producing an antibody, comprising:

(a) immunizing an animal with a Δ

5-desaturase polypeptide fragment derived from fungus or algae; and

(b) isolating the antibody therefrom.

18. The method of claim 17 wherein the animal is immunized with a Δ

5-desaturase polypeptide fragment derived from Saprolegnia diclina or Mortierella alpina.

19. The method of claim 18 wherein the animal is immunized with a Δ

5-desaturase polypeptide fragment comprising at least 8 contiguous amino acid residues of SEQ ID NO;

2 or conservative variations of SEQ ID NO;

20. The method of claim 19 wherein the animal is immunized with a Δ

5-desaturase polypeptide fragment comprising SEQ ID NO;

2 or conservative variations of SEQ ID NO;

21. A method of detecting Δ

5-desaturase in a sample comprising;

(a) exposing the sample to an antibody that specifically binds Δ

5-desaturase derived from fungus or algae, wherein the exposing is performed under conditions that allow binding of the antibody to Δ

5-desaturase; and

(b) detecting a bound antibody.

22. The method of claim 21 further comprising:

(c) quantitating the bound antibody from step (b).

23. The method of claim 21 wherein the sample is exposed to an antibody that specifically binds a Δ

5-desaturase polypeptide fragment derived from fungus or algae and the exposing is performed under conditions that allow binding of the antibody to the Δ

5-desaturase polypeptide fragment.

24. The method of claim 23 wherein the sample is exposed to an antibody that specifically binds a Δ

5-desaturase polypeptide fragment derived from Saprolegnia diclina or Mortierella alpina and the exposing is performed under conditions that allow binding of the antibody to the Δ

5-desaturase polypeptide fragment.

25. The method of claim 24 wherein the sample is exposed to an antibody that specifically binds a polypeptide fragment comprising at least 8 contiguous amino acid residues of SEQ ID NO:

2 or conservative variations of SEQ ID NO;

2 and the exposing is performed under conditions that allow binding of the antibody to the Δ

5-desaturase polypeptide fragment.

26. The method of claim 25 wherein the sample is exposed to an antibody that specifically binds a polypeptide fragment comprising SEQ ID NO:

2 or conservative variations of SEQ ID NO;

2 and the exposing is performed under conditions that allow binding of the antibody to the Δ

5-desaturase polypeptide fragment.

27. The method of claim 21 wherein the sample is derived from a host that natively expresses Δ

5-desaturase.

28. The method of claim 21 wherein the sample is derived from a host suspected of expressing a heterologous Δ

5-desaturase.

29. The method of claim 21 wherein the sample comprises a seed extract.

30. The method of claim 29 wherein the seed extract is derived from a transgenic seed suspected of expressing Δ

5-desaturase.

31. An immunogenic isolated polypeptide consisting essentially of SEQ ID NO:

2 or conservative variations of SEQ ID NO;

32. An isolated nucleic acid having a sequence that encodes an immunogenic polypeptide consisting essentially of SEQ ID NO:

2 or conservative variations of SEQ ID NO;
