Abstract

Methods, compositions and kits are disclosed. The methods are directed to determining the presence or relative amounts of two or more components in a medium. A combination is provided comprising a medium suspected of containing the components, at least two sensitizer reagents and at least one reactive reagent activatable by singlet oxygen. The sensitizer reagents are capable of generating singlet oxygen and are distinguishable by wavelength of sensitization. The combination of sensitizer reagents and reactive reagents allows differential detection of the components. The sensitizer reagents are differentially activated. The amount of signal generated as a result of the activation of said reactive reagent is determined wherein the amount thereof is related to the amount of each of the components in the medium.

Claims

1. A method for determining the presence or relative amounts of two or more components in a medium, said method comprising:

a) providing in combination (1) a medium suspected of containing said components, (2) at least two sensitizer reagents, said sensitizer reagents being capable of generating a product upon activation, said sensitizer reagents being distinguishable on the basis of activation, and (3) at least one reactive reagent activatable by said product, wherein the combination of sensitizer reagent and reactive reagent allows differential detection of said components, and

b) differentially activating said sensitizer reagents and detecting the amount of signal generated corresponding to the activation of said reactive reagent by said product, the amount thereof being related to the amount of each of said components in said medium.

2. A method for determining the presence or relative amounts of two or more components in a medium, said method comprising:

a) providing in combination (1) a medium suspected of containing said components, (2) at least two sensitizer reagents, said sensitizer reagents being capable of generating singlet oxygen and being distinguishable on the basis of activation, and (3) at least one reactive reagent activatable by singlet oxygen, wherein the combination of sensitizer reagents and reactive reagents allows differential detection of said components, and

b) differentially activating said sensitizer reagents and detecting the amount of signal generated as a result of the activation of said reactive reagent, the amount thereof being related to the amount of each of said components in said medium.

3. A method according to claim 2 wherein said reactive reagent is a chemiluminescent composition.

4. A method according to claim 2 wherein multiple reactive reagents are employed.

A method according to claim 2 wherein one of said sensitizer reagents or said reactive reagent is associated with a first specific binding pair (sbp) member and wherein said first sbp member is capable of binding to said component or to a second sbp member to form a complex related to the amount of said component.

6. A method according to claim 5 wherein said first sbp member is capable of binding to said component and wherein a second sbp member that is associated with the other of said sensitizer reagents or said reactive reagent is capable of binding with said component.

7. A method for determining the presence or relative amounts of two or more components in a medium, said method comprising:

a) providing in combination (1) a medium suspected of containing said components, (2) at least two sensitizer reagents, said sensitizer reagents being capable of generating singlet oxygen and being distinguishable by wavelength of sensitization, and (3) at least one reactive reagent activatable by singlet oxygen wherein a first specific binding pair (sbp) member is associated with each of said sensitizer reagents and wherein said first sbp member is capable of binding to said component or to a second sbp member to form a complex related to the amount of said component, and

b) differentially activating said sensitizer reagents and detecting the amount of signal generated as a result of the activation of each of said reactive reagents, the amount thereof being related to the amount of each of said components in said medium.

8. A method according to claim 7 wherein multiple reactive reagents are employed, said reactive reagents being differentially detectable.

9. A method according to claim 8 wherein said multiple reactive reagents are chemiluminescent compositions that are differentially detectable by different wavelengths of emission or by different rates of decay.

10. A method according to claim 7 wherein said second sbp member is capable of binding with said component.

11. A method according to claim 7 wherein at least one of said reactive reagents comprises a fluorescent energy acceptor,

12. A method according to claim 7 wherein at least one of said reactive reagents is photoactive indicator precursor that is activated by singlet oxygen to form a photoactive indicator.

13. A method according to claim 7 wherein said differentially activating comprises irradiating said medium with light of different wavelengths.

14. A method according to claim 7 wherein said reactive reagents are activatable by reaction with singlet oxygen.

15. A method according to claim 7 wherein at least one of said sensitizers is a photosensitizer.

16. A method according to claim 15 wherein said photosensitizer is a dye.

17. A method according to claim 7 wherein at least one of said reactive reagents or said sensitizer reagents is associated with a particle.

19. A method according to claim 17 wherein at least one of said photosensitizers is a dye.

20. A method according to claim 17 wherein at least one of said photosensitizers is selected from the group consisting of naphthocyanines, phthalocyanines, thiazines, porphyrins, metallo-porphyrins, oxazines, cyanines, squaraines, xanthenes, merocyanines

21. A method according to claim 17 wherein at least one of said chemiluminescent compositions is an olefinic compound.

22. A method according to claim 20 wherein said olefinic compound is selected from the group consisting of thioxenes, dihydrooxazines and dioxenes.

23. A method according to claim 17 wherein said first and said second particles are independently selected from the group consisting of latex particles, liposomes and oil droplets.

24. A method according to claim 17 wherein said components are selected from the group consisting of ligands, receptors and polynucleotides.

18. A homogeneous method for determining the presence or relative amounts of two or more components in a medium, said method comprising:

a) providing in combination (1) a medium suspected of containing said components, (2) a photosensitizer for each of said components, said photosensitizer being associated with a first particle and being capable of generating singlet oxygen and having different wavelengths of sensitization, and (3) chemiluminescent compositions activatable by singlet oxygen, each of which is associated with a second particle, wherein one or more of said chemiluminescent compositions are differentially detectable by different wavelengths of emission or by different rates of decay and wherein the different wavelengths of sensitization and the different wavelengths of emission or different rates of decay result in the differential detection of each of said components and wherein a first specific binding pair (sbp) member is associated with each of said photosensitizers and wherein said first sbp member is capable of binding to said component or to a second sbp member to form a complex related to the amount of said component, and

b) differentially irradiating said photosensitizer with light and detecting the amount of luminescence generated by each of said chemiluminescent compositions at a time after activation and at a wavelength corresponding to the rate of decay and wavelength of said luminescent emission, the amount thereof being related to the amount of each of said components in said medium.

25. A kit comprising in packaged combination:

(a) a plurality of sensitizer reagents, each comprising (1) a sensitizer composition that is capable of generating singlet oxygen and (2) a member of a specific binding pair (sbp), wherein at least a portion of said sensitizer compositions have different wavelengths of sensitization, and

b) at least one reactive reagent that is activatable by singlet oxygen.

26. A kit according to claim 25 comprising multiple reactive reagents.

27. A kit according to claim 26 wherein one or more of said multiple reactive reagents are differentially detectable.

28. A kit according to claim 27 wherein said multiple reactive reagents are chemiluminescent compositions that are differentially detectable by different wavelengths of emission or by different rates of decay.

29. A kit according to claim 25 wherein said reactive reagent is associated with an sbp member that is capable of binding with said component.

30. A kit according to claim 25 wherein at least one of said reactive reagents is photoactive indicator precursor that is activated by singlet oxygen to form a photoactive indicator.

31. A kit according to claim 25 wherein at least one of said sensitizers is a photosensitizer.

32. A kit according to claim 31 wherein said photosensitizer is a dye.

33. A kit according to claim 25 wherein at least one of said reactive reagents or one of said sensitizers is associated with a particle.

34. A compound which is bis(tri-alkyl¹-silyl)silicon tetra-alkyl²-naphthalocyanine.

35. A compound according to claim 34 wherein alkyl¹ is hexyl.

36. A compound according to claim 34 wherein alkyl² is butyl.

37. A compound according to claim 34 which is bis(tri-n-hexylsilyl)silicon tetra-t-butyl-naphthalocyanine.

38. A compound which is dihydroxysilicon tetra-t-butyl-naphthalocyanine or dichlorosilicon tetra-t-butyl-naphthalocyanine.

39. A method of making bis(tri-n-hexylsilyl)silicon tetra-t-butyl-naphthalocyanine, said method comprising treating dihydroxysilicon tetra-t-butyl-naphthalocyanine with tri-n-hexylsilylchloride.
