Abstract

Provided is a preservation solution for preserving biological material at low temperature including one or more polyphenols and a method for preservation of biological material, the method includes adding the preservation solution to biological material, cooling the biological material and storing it under appropriate storing conditions. The present method may be used for hypothermic preservation or for cryopreservation, including freezing and lyophilization, and may be used with any biological material, including cells selected from RBC, WBC, MNC, UCB MNC and bacteria. In the case of RBC, also disclosed is a method for its freezing such that upon thawing, the material has less than 2% free hemoglobin.

54. A method for preservation of a biological material selected from the group consisting of cells, cell aggregates, tissue, organs, natural liposomes, and synthetic liposomes, the method comprising:

(a) adding a preservation solution to the biological material, the preservation solution comprising one or more polyphenols;

(b) freeze drying the biological material to produce a frozen biological material; and

(c) storing the frozen biological material under appropriate storing conditions.

55. The method of claim 54, wherein the polyphenols comprise one or more catechins.

56. The method of claim 55, wherein the catechin is epigallocatechin gallate (EGCG).

57. The method of claim 54, wherein the polyphenols are derived from green tea extract (GTE).

58. The method of claim 54, wherein the preservation solution does not comprise a significant amount of glycerol.

59. The method of claim 54, wherein the preservation solution does not comprise a significant amount of DMSO.

60. The method of claim 54, wherein the preservation solution comprises a macromolecule.

61. The method of claim 54, wherein the preservation is cryopreservation, the preservation solution is a cryopreservation solution and the freeze drying of step (c) is to a temperature below 0°

C.

62. The method of claim 54, wherein the biological material comprises cells selected from the group consisting of red blood cells (RBC), white blood cells (WBC), mononuclear cells (MNC), umbilical cord blood cells (UCB), hematopoietic stem cells (HSC), and bacteria.

63. The method of claim 62, wherein the biological material comprises RBC and the frozen biological material is such that after thawing under appropriate thawing conditions the biological material comprises free hemoglobin levels below 10 percent.

65. The method of claim 54, wherein the one or more polyphenols are present in the preservation solution at a concentration of from about 50 M to about 4 mM.

64. A method for preservation of a biological material selected from the group consisting of cells, cell aggregates, tissue, organs, natural liposomes, and synthetic liposomes, the method comprising:

(a) adding to the biological material a preservation solution comprising one or more polyphenols and essentially free of any polyalcohol;

(b) freeze drying the biological material; and

(c) storing the biological material under appropriate storing conditions.

66. The method of claim 64, wherein the one or more polyphenols are present in the preservation solution at a concentration of from about 50 M to about 4 mM.
