Abstract

The present invention provides microfabricated substrates and methods of conducting reactions within these substrates. The reactions occur in plugs transported in the flow of a carrier-fluid.

Claims

1. A microfluidic system comprising:

a substrate comprising one or more inlet ports, one or more outlet ports, and an array of traps in fluidic communication with the inlet and outlet ports,

one or more means for applying force to introduce a fluid via the one or more inlet ports so that a plurality of fluid portions are trapped in the array of traps;

wherein each fluid portion comprises reagents sufficient for an autocatalytic reaction including a first species of molecule in a concentration such that each fluid portion contains no more than a single molecule of the first species, and wherein the fluid portions are trapped at least for a period of time sufficient for the autocatalytic reaction to occur such that the single molecule is amplified.

2. The system of claim 1, wherein the fluid portions are moved to the one or more outlet ports after the period of time has passed.

3. The system of claim 2, wherein one or more of the outlet ports is also an inlet port.

4. The system of claim 1, wherein the single molecule is a single biological molecule.

5. The system of claim 4, wherein the single biological molecule is DNA or RNA and the autocatalytic reaction is a polymerase-chain reaction.

6. The system of claim 1, further comprising a detector to detect, monitor, or analyze one or more properties of the autocatalytic reaction during and/or after it has occurred.

7. The system of claim 6, wherein the properties comprise light emission.

8. The system of claim 7, wherein the reagents include one or more fluorescent labels and the detector detects emissions from the one or more fluorescent labels.

9. The system of claim 1, wherein the fluid portions are trapped for a period of time that may range from tens of microseconds to hours to weeks.

10. The system of claim 1, wherein the array of traps comprises an array of connected microchannels.

11. The system of claim 10, further comprising one or more valves that are used to control the flow of fluid portions into and/or out of the traps.

12. The system of claim 1, wherein each fluid portion is a fluid plug.

13. The system of claim 12, wherein each fluid plug is substantially spherical in shape.

14. The system of claim 12, wherein each fluid plug is substantially surrounded by and immiscible in a carrier fluid.

15. The system of claim 1, wherein the means for applying force is a means for applying pressure.
