Abstract

The present disclosure relates to an LNA oligonucleotide consisting of a sequence selected from the group consisting of 5′-(T_x)G_xG_xc_sa_sa_sg_sc_sa_st_sc_sc_sT_xG_xT-3′ and 5′-(G_x)T_xa_sc_st_sg_sc_sc_st_st_sc_sT_xT_xA-3′, wherein capital letters designate a beta-D-oxy-LNA nucleotide analogue, small letters designate a 2-deoxynucleotide, underline designates either a beta-D-oxy-LNA nucleotide analogue or a 2-deoxynucleotide, subscript “s” designates a phosphorothioate link between neighbouring nucleotides/LNA nucleotide analogues, and subscript “x” designates either a phosphorothioate link or a phosphorodiester link between neighbouring nucleotides/LNA nucleotide analogues, and wherein the sequence is optionally extended by up to five 2-deoxynucleotide units. The LNA oligonucleotides are useful for modulating the expression of hypoxia-inducible factor-1a (HIF-1a), e.g. in the treatment of cancer diseases, inhibiting angiogenesis, inducing apoptosis, preventing cellular proliferation, or treating an angiogenic disease, e.g. diabetic retinopathy, macular degeneration (ARMD), psoriasis, rheumatoid arthritis and other inflammatory diseases.

31. A method for treating cancer, said method comprising administering a composition comprising a therapeutically effective amount of an LNA oligonucleotide consisting of the sequence:

32. The method according to claim 31, wherein the cancer is a solid tumor.

33. The method according to claim 31, wherein the cancer is selected from the group consisting of multiple myeloma, renal cancer, cervical cancer, colon cancer, brain cancer, and breast cancer.

34. The method of claim 31, wherein the agent is a chemotherapeutic agent.

35. The method of claim 34, wherein the chemotherapeutic agent is selected from the group consisting of doxorubicin, vincristine, dexamethasone, prednisone and a taxane compound.

36. The method of claim 31, wherein the composition is administered parenterally.

37. The method of claim 31, wherein the composition is administered intravenously.

38. The method of claim 31, wherein the composition is administered by bolus injection into a target organ.

39. The method of claim 31, wherein the composition is administered intraperitoneally.
