Abstract

Compositions and methods of producing vaccines, including methods wherein whole organism vaccines are provided with limited replication abilities, thereby increasing vaccine safety and efficacy, through the use of non-natural, unnatural, or non-naturally encoded amino acids.

Claims

1. A mycobacterium avium subspecies paratuberculosis (MAP) cell wherein a non-natural amino acid has been incorporated into the amino acid sequence of an essential gene product.

2. The MAP of claim 1, wherein the non-natural amino acid is incorporated into a MAP gene product that is required for replication.

3. The MAP of claim 1 wherein more than one non-natural amino acid is incorporated into more than one essential gene product.

4. An E. coli cell wherein a non-natural amino acid has been incorporated into an essential gene product.

5. A bacterial cell wherein a non-natural amino acid has been incorporated into an essential gene product.

6. A cell wherein a non-natural amino acid has been incorporated into an essential gene product.
