Abstract

Disclosed herein are methods for identifying inhibitors of the catalytic activity of LOXL2 using in vitro assays. In certain embodiments, inhibitors identified by these in vitro assays are effective in inhibiting tumor desmoplasia and fibrosis.

Claims

1. A method for identifying an inhibitor of LOXL2 activity, the method comprising:

(a) providing a co-culture of endothelial cells growing in the presence of stromal cells;

(b) adding a test molecule to the co-culture; and

(c) assaying the co-culture for angiogenesis or vasculogenesis;

wherein a test molecule that reduces the degree of angiogenesis or vasculogenesis in the co-culture, compared to a co-culture in the absence of the test molecule, is identified as an inhibitor of LOXL2 activity.

2. The method of claim 1, wherein the endothelial cells are human endothelial cells.

3. The method of claim 2, wherein the endothelial cells are human umbilical vein endothelial cells (HUVEC).

4. The method of claim 1, wherein the stromal cells are human cells.

5. The method of claim 1, wherein the stromal cells are fibroblasts.

6. The method of claim 1, wherein the test molecule is a polypeptide.

7. The method of claim 6, wherein the polypeptide is an antibody.

8. The method of claim 7, wherein the antibody is an anti-LOXL2 antibody.

9. The method of claim 1, wherein the test molecule is a nucleic acid.

10. The method of claim 9, wherein the nucleic acid is a siRNA.

11. The method of claim 1, wherein the test molecule is a small organic molecule with a molecular weight less than 1,000 D.

12. The method of claim 1, wherein reduction in the degree of angiogenesis or vasculogenesis is indicated by a reduction in vessel number or density.

13. The method of claim 1, wherein reduction in the degree of angiogenesis or vasculogenesis is indicated by a reduction in vessel length.

14. The method of claim 1, wherein reduction in the degree of angiogenesis or vasculogenesis is indicated by a reduction in the degree of vessel branching.

15. The method of claim 1, wherein reduction in the degree of angiogenesis or vasculogenesis is indicated by a reduction in levels of CD31.
