Abstract

Disclosed herein are compounds that form covalent bonds with Bruton'"'"'s tyrosine kinase (Btk). Also described are irreversible inhibitors of Btk. Methods for the preparation of the compounds are disclosed. Also disclosed are pharmaceutical compositions that include the compounds. Methods of using the Btk inhibitors are disclosed, alone or in combination with other therapeutic agents, for the treatment of autoimmune diseases or conditions, heteroimmune diseases or conditions, cancer, including lymphoma, and inflammatory diseases or conditions.

22. A method for treating a cancer in a subject in need thereof comprising administering to the subject two or more agents, wherein the first agent is a selective, covalent inhibitor of Bruton'"'"'s tyrosine kinase (Btk) and the second agent is an anti-cancer drug.

23. The method of claim 22, wherein the cancer is a lymphoma.

24. The method of claim 22, wherein the cancer is selected from nodal marginal zone B cell lymphoma, extranodal marginal zone B cell lymphoma, splenic marginal zone B cell lymphoma, diffuse large B cell lymphoma, follicular lymphoma, Waldenstrom macroglobulinemia, mantle cell lymphoma, or burkitt lymphoma.

25. The method of claim 22, wherein the cancer is plasma cell myeloma.

26. The method of claim 22, wherein the cancer is a leukemia.

27. The method of claim 22, wherein the cancer is selected from chronic lymphocytic leukemia, B-cell prolymphocytic leukemia, or Burkitt leukemia.

28. The method of claim 22, wherein the cancer is a solid tumor.

29. The method of claim 22, wherein the anti-cancer drug is a proapoptotic agent, angiogenesis inhibitor, dexamethasone, or a combination thereof.

30. The method of claim 22, wherein the anti-cancer drug is a proapoptotic agent.

31. The method of claim 22, wherein the anti-cancer drug is an angiogenesis inhibitor.

32. The method of claim 22, wherein the anti-cancer drug is dexamethasone.

33. The method of claim 22, wherein the selective, covalent inhibitor of Btk and the anti-cancer drug are administered simultaneously.

34. The method of claim 22, wherein the selective, covalent inhibitor of Btk and the anti-cancer drug are administered sequentially.

35. The method of claim 22, wherein the selective, covalent inhibitor of Btk is administered orally.

36. The method of claim 22, wherein the selective, covalent inhibitor of Btk is an irreversible Btk inhibitor.

37. The method of claim 22, wherein the selective, covalent inhibitor of Btk targets a cysteine residue of a Bruton'"'"'s tyrosine kinase (Btk).

38. The method of claim 37, wherein the covalent bond is formed between a portion of a Michael acceptor moiety on the inhibitor of Btk and a portion of a cysteine residue of a Bruton'"'"'s tyrosine kinase (Btk).

39. The method of claim 38, wherein the Michael acceptor moiety is an acrylamide, vinyl sulfonamide or propargylamide.

40. The method of claim 22, wherein the covalent bond is formed from a functional group on the Btk inhibitor and the functional group is capable of covalently binding a cysteine residue of a Bruton'"'"'s tyrosine kinase (Btk).

41. The method of claim 40, wherein the cysteine residue is cysteine 481 of a Bruton'"'"'s tyrosine kinase (Btk).

42. A method for treating a cancer in a subject in need thereof comprising administering to the subject two or more agents, wherein the first agent is a selective, covalent inhibitor of Bruton'"'"'s tyrosine kinase (Btk) and the second agent is an agent for treating plasma cell myeloma.
