Abstract

The present invention provides novel microfluidic substrates and methods that are useful for performing biological, chemical and diagnostic assays. The substrates can include a plurality of electrically addressable, channel bearing fluidic modules integrally arranged such that a continuous channel is provided for flow of immiscible fluids.

Claims

75. A method for analyzing a reaction product, the method comprising:

providing a plurality of droplets in a reservoir, at least one of the droplets comprising molecules for a reaction;

conducting a reaction in the at least one droplets; and

moving the droplets through a channel and past a detector, thereby detecting a reaction product in at least one of the at least one droplets.

76. The method according to claim 75, wherein the reaction is conducted in the reservoir.

77. The method according to claim 75, wherein the droplets are separated by an immiscible fluid.

78. The method accordingly to claim 77, wherein the droplets and the immiscible fluid have different buoyancies.

79. The method according to claim 78, wherein the buoyancy differences assist droplet collection by separating at least a portion of the immiscible fluid from the droplets.

80. The method according to claim 75, wherein the droplets are monodisperse within the channel.

81. The method according to claim 77, wherein the immiscible fluid is an oil

82. The method according to claim 81, wherein the oil is a fluorinated oil.

83. The method according to claim 81, wherein the oil comprises a surfactant.

84. The method according to claim 83, wherein the surfactant the surfactant is a fluorosurfactant.

85. The method according to claim 75, wherein the reaction is a temperature controlled reaction.

86. The method according to claim 75, wherein the molecules comprise one or more cells, virus particles, bacteria, beads, protein molecules, nucleic acid molecules, or any combination thereof.

87. The method according to claim 86, wherein the molecules further comprise reagents for an amplification reaction.

88. The method according to claim 87, wherein the amplification reaction is the polymerase chain reaction.

89. The method according to claim 75, wherein after detecting, the method further comprising sorting the droplets.

90. The method according to claim 89, further comprising collecting at least one of the sorted droplets.

91. The method according to claim 75, wherein the detector is an electrical capacity detector or an optical detector.

92. The method according to claim 91, wherein the optical detector comprises a laser.

93. The method according to claim 75, wherein the moving step further comprises spacing the droplets.

94. The method according to claim 93, wherein spacing comprises adding an immiscible fluid to the channel so as to increase spacing between droplets prior to the droplets moving past the detector.

95. The method according to claim 75, wherein the droplets pass through the channel and are detected by the detector one at a time.

96. The method according to claim 95, wherein the width of the channel is selected and arranged to position the droplets so that they are detected by the detector one at a time.

97. The method according to claim 75, wherein the at least one droplet is a merged, fused, or coalesced droplet.

98. The method according to claim 75, further comprising the step of merging a reagent into the droplets prior to detecting the reaction product.

99. The method according to claim 86, wherein the molecule is a protein, and the reaction is an enzymatic reaction.

100. The method according to claim 75, wherein the reservoir is an off-chip reservoir.

101. The method according to claim 75, wherein moving includes by aspiration.

102. The method according to claim 75, wherein providing includes by aspiration.
