Abstract

The invention relates to methods and devices for preparing synthetic nucleic acids.

Claims

1. A method of preparation of synthetic nucleic acids, in particular of nucleic acid double strands, comprising the steps:

a. preparation of a multiplicity of different nucleic acid fragments by solid-phase synthesis, preferably at various positions of a common solid support, and

b. joining together of at least two nucleic acid fragments from (a) by binding to one another or by covalent linkage,

characterized in that at least some nucleic acid fragments, which have a high AT content, are used in an increased amount relative to other fragments for step (b).

2. The method as claimed in claim 1 characterized in that a copy of the synthetic nucleic acids is provided by an RNA polymerase.

3. The method as claimed in claim 2 characterized in that the nucleic acid fragments produced are used for the modulation of gene expression by RNAi and/or antisense methods.

4. The method as claimed in claim 2 characterized in that the nucleic acid fragments produced are used for the production, extraction, purification, isolation and/or preparation of analytes and/or samples (sample preparation).

A polypeptide with defined properties for use as a pharmaceutically active substance and/or for use in the production of a pharmaceutically active substance, produced by synthesis of synthetic nucleic acids with the sequence encoding the polypeptide, in particular of nucleic acid double strands, comprising the steps:

a. preparation of a multiplicity of different nucleic acid fragments by solid-phase synthesis, preferably at various positions of a common solid support,

b. joining together of at least two nucleic acid fragments from (a) by binding to one another,

characterized in that at least some nucleic acid fragments, which have a high AT content, are used in an increased amount relative to other fragments for step (b), and where the method further comprises;

c. production of the polypeptide using said sequence.

6. A method of production of a polypeptide, comprising steps (a), (b) and (c) from claim 5.
