Abstract

The present disclosure provides devices and uses thereof A devices disclosed herein comprises a plurality of microneedles adapted to protrude from the device. In some embodiments, a device is dimensioned and constructed to carry a payload, so that the payload can be delivered to an internal tissue of a subject or through a wall of a vessel after interaction with microneedles. In some embodiments, devices can be used for oral or intravenous administration. In some embodiments, devices can be used for implantation such as vaginal, rectal, urethral or bladder suppository or pessary.

Claims

1. A device dimensioned for administration or implantation to an internal tissue of a subject, which device includes a plurality of microneedles adapted to protrude from the device surface so that the microneedles interact with the internal tissue.

2. The device of claim 1, wherein the device is dimensioned for oral, sublingual or intraoperative administration.

3. The device of claim 1, wherein the device is dimensioned for intravenous or respiratory administration.

4. The device of claim 3, wherein the internal tissue is a part of a vessel wall.

5. The device of claim 1, wherein the device is dimensioned for vaginal, rectal, urethral or bladder suppository or pessary.

6. The device of any claim 1. being dimensioned and constructed to carry a payload, which payload is delivered to the internal tissue after interaction with the microneedles.

7. The device of claim 1, wherein a target site of the subject is a viscus.

10. The device of claim 1, wherein a target site of the subject is in an internal GI tract.

12. The device of claim 1, wherein some or all of the microneedles are hollow.

13. The device of claim 12, wherein at least one of the hollow microneedles is cannulated, defining a channel that enables storage or introduction of the payload from the other parts of the device.

14. The device of claim 1, wherein the device has a lumen bounded by a wall.

16. The device of claim 1, wherein the microneedles of the plurality protrude substantially radially.

25. The device of claim 1, wherein each microneedle has a length within a range of about 1 micron and about 2 cm.

26. The device of claim 1, wherein a portion of the device is or comprises a magnetic material.

29. The device of claim 1, wherein at least one microneedle is or comprises a degradable material.

34. The device of claim 1, further comprising a coating.

The device of claim 34, wherein the coating is selected from a group consisting of a pH-sensitive coating, an inflammatory-sensitive coating, a virus/bacteria-sensitive coating, a cancer specific coating, a hydrolytically degradable coating, a medicated coating, an inert coating and any combination thereof.

39. The device of claim 1. arranged and constructed to transition from an initial state to an exposed state that differ from one another in extent to which microneedles in the plurality protrude from the device surface.

53. The device of claim 52, wherein the trigger is selected from the group consisting of presence of blood, specific molecules/entities, magnetic fields, ultrasound, electric fields, pH, enzymatic activity, temperature, light, mechanical forces and any combination thereof.

57. The device of claim 1, wherein the payload is or comprises an agent selected from the group consisting of a therapeutic agent, a prophylactic agent, a nutraceutical agent, a diagnostic agent or any combination thereof.

60. A method of delivering a payload, comprising:

providing a device dimensioned for administration or implantation, wherein the device includes a plurality of microneedles adapted to protrude from the device surface, which device comprises a payload,

administering or implanting the device to a subject; and

releasing the payload to an internal tissue of the subject after interaction with the microneedles.
