Abstract

The invention relates to a method of manufacturing a scaffold for tissue engineering or repair. The method includes providing a mixture comprising a degradable hydrogel, a degradable particle, and living cells. The mixture is then incubated under conditions which allow proliferation of the living cells and degradation of the degradable particle in the hydrogel. The degradable hydrogel of the incubated mixture is then degraded at least partially, followed by adding further living cells to obtain the scaffold.

Claims

1. A method of manufacturing a scaffold for tissue engineering or repair, comprising:

providing a mixture comprising at least one degradable hydrogel, at least one kind of degradable particle and one or more species of living cells to form a hydrogel comprising the at least one kind of degradable particle and the one or more species of living cells, wherein the degradable particle comprises a material which degrades faster than the degradable hydrogel;

incubating the mixture under conditions which allow proliferation of the one or more species of living cells and degradation of the at least one kind of degradable particle in the hydrogel to allow the at least one kind of degradable particle to degrade and to allow the one or more species of living cells to proliferate;

degrading at least partially the at least one degradable hydrogel of the incubated mixture; and

adding one or more species of living cells after degrading at least partially the at least one degradable hydrogel of the incubated mixture, wherein the one or more species of living cells added after degrading at least partially the at least one degradable hydrogel of the incubated mixture comprises the same or different species of cells than the cells already mixed into the mixture together with the at least one degradable hydrogel and the at least one kind of degradable particle to obtain the scaffold.

2. The method of claim 1, further comprising the step of:

(v) incubating the scaffold under conditions which allow proliferation of the one or more species of living cells added after degrading at least partially the at least one degradable hydrogel of the incubated mixture.

3. The method of claim 2, wherein the proliferation of the one or more species of living cells added after degrading at least partially the at least one degradable hydrogel of the incubated mixture occurs at voids formed by the degraded at least one degradable hydrogel.

4. The method of claim 1, wherein the at least one degradable hydrogel comprises a natural polymer selected from the group consisting of polysaccharide, glycosaminoglycan, protein, and a mixture thereof.

5. The method of claim 4, wherein the polysaccharide is selected from the group consisting of alginate, agarose, chitosan, dextran, starch, gellan gum, and a mixture thereof.

6. The method of claim 5, wherein the at least one degradable hydrogel comprises alginate.

The method of claim 1, wherein the one or more species of living cells already mixed into the mixture together with the at least one degradable hydrogel and the at least one kind of degradable particle, or the one or more species of living cells added after degrading at least partially the at least one degradable hydrogel of the incubated mixture is selected from the group consisting of osteogenic cells, fibroblasts, epidermal cells, adipocytes, neural cells, endothelial cells, epithelial cells, keratinocytes, hepatocytes, myocytes, cells from joint ligaments, cells from the nucleus pulposis, chondrocytes, embryonic stem cells, adult stem cells, endodermal lineage cells, carcinoma cells used for regenerative medicine, and a mixture thereof.

The method of claim 7, wherein the one or more species of living cells already mixed into the mixture together with the at least one degradable hydrogel and the at least one kind of degradable particle, or the one or more species of living cells added after degrading at least partially the at least one degradable hydrogel of the incubated mixture comprises chondrocytes.

9. The method of claim 1, wherein the at least one kind of degradable particle comprises a degradable material which degrades at body temperature.

10. The method of claim 1, wherein the at least one kind of degradable particle comprises a degradable material which is selected from the group consisting of hydrocarbon, hydrogel and organic compound.

11. The method of claim 10, wherein the at least one kind of degradable particle is surface cross-linked gelatin.

12. The method of claim 1, wherein the at least one degradable hydrogel or the at least one kind of degradable particle further comprises at least one kind of biologically active molecule.

13. The method of claim 12, wherein the at least one kind of biologically active molecule is selected from the group consisting of growth factor, cytokine, antibiotic, anti-inflammatory agent, analgesic, a cell attachment molecule, and a mixture thereof.

14. A method of repairing a damaged tissue, comprising:

providing a scaffold obtainable by a method of claim 1; and

contacting the scaffold with a damaged tissue of a subject in need thereof.

15. The method of repairing a damaged tissue of claim 14, wherein the contacting comprises implanting the scaffold at the damaged tissue site or the surrounding of the damaged tissue site.
