Abstract

The present invention is directed to fusion proteins having an IL-21 cytokine portion and an anti-CD20 antibody portion, and methods of using such fusion proteins. The invention also provides pharmaceutical compositions and kits utilizing the IL-21-anti-CD20 fusion proteins. In particular, the methods, kits and compositions of the invention are useful in the treatment of cancer and autoimmune diseases.

Claims

1. A recombinant fusion protein comprising a first amino acid sequence for IL-21 or functional fragment thereof, and a second amino acid sequence for anti-CD20 antibody or functional fragment thereof.

2. The fusion protein of claim 1 further comprising a linker peptide between said first amino acid sequence and said second amino acid sequence.

3. The fusion protein of claim 1 wherein said first amino acid sequence is operatively connected to an N-terminal end of said second amino acid sequence.

4. The fusion protein of claim 3 comprising an amino acid sequence as set forth in SEQ ID NO:

5. The fusion protein of claim 3 further comprising a linker peptide of SEQ ID NO:

1 between said first amino acid sequence and said second amino acid sequence.

6. The fusion protein of claim 1 wherein said first amino acid sequence is operatively connected to a C-terminal end of said second amino acid sequence.

7. The fusion protein of claim 1 wherein said functional fragment of anti-CD20 antibody specifically binds one or more epitope comprising the amino acid sequence set forth in SEQ ID NO:

2 or SEQ ID NO;

3, or combinations thereof.

8. The fusion protein of claim 1 wherein said functional fragment of anti-CD20 antibody specifically binds a polypeptide having an amino acid sequence as set forth in SEQ ID NO:

4, or portions thereof.

9. The fusion protein of claim 1, wherein said amino acid sequence for anti-CD20 antibody is a monoclonal antibody.

10. The fusion protein of claim 1, wherein said anti-CD20 antibody fragment is selected from the group consisting of Fab, Fab′

, F(ab′

)2, and Fv fragments, diabodies, single chain antibody molecules, and multispecific antibodies formed from antibody fragments.

11. The fusion protein of claim 1 further comprising a label.

13. An isolated polynucleotide comprising a sequence that encodes the fusion protein of claim 1.

16. An expression vector encoding the fusion protein of claim 1.

17. A cultured cell expressing the fusion protein of claim 1.

18. A pharmaceutical composition comprising the fusion protein of claim 1 and at least one pharmaceutically acceptable carrier.

19. The pharmaceutical composition of claim 18, further comprising one or more pharmaceutically acceptable excipient, additive, or adjuvant.

20. The pharmaceutical composition of claim 18 further comprising at least one of a chemotherapeutic agent, a radioactive isotope, a therapeutic agent, or combinations thereof.

21. A method of treating a disorder in a subject comprising administering a therapeutically effective amount of the pharmaceutical composition of claim 18 to the subject.

22. A kit comprising the pharmaceutical composition of claim 18.

23. A method of treating a disorder in a subject comprising administering a therapeutically effective amount of the recombinant fusion protein of claim 1 to the subject.

24. The method of claim 23 wherein the disorder is a cell-proliferative disorder.

25. The method of claim 24 wherein the cell-proliferative disorder is cancer.

26. The method of claim 25 wherein the cancer is lymphoma.

27. The method of claim 23 wherein the disorder is an autoimmune disease.

28. The method of claim 23 wherein the subject is a human.

29. The method of claim 23 further comprising administering to the subject a therapeutically effective amount of at least one agent selected from the group consisting of a chemotherapeutic agent, a radioactive isotope, a therapeutic agent, or combinations thereof.

30. The method of claim 23 wherein the fusion protein is administered with a pharmaceutically acceptable carrier.

31. A kit comprising the recombinant fusion protein of claim 1.

32. The kit of claim 31 further comprising at least one agent capable of facilitating detection of said fusion protein.

33. A method of targeted delivery to one or more tumors in a subject comprising administering a therapeutically effective amount of the fusion protein of claim 1 to the subject.

34. The method of claim 33 wherein the one or more tumors express CD20.

35. The method of claim 33 further comprising activating one or more immune effector cells adjacent the one or more tumors by the recombinant fusion protein.

36. A method of inhibiting the growth of a tumor cell comprising contacting the tumor cell with the recombinant fusion protein of claim 1.

37. A method of targeting multiple receptors on a cell with a single protein, comprising contacting the cell with the recombinant fusion protein of claim 1.

38. A method of enhancing antibody dependent cellular cytotoxicity activity against cells expressing CD20 in a subject comprising administering the recombinant fusion protein of claim 1 to the subject.

12. The fusion protein of claim 12, wherein said label is a radiolabel.

14. A cloning vector comprising the nucleotide sequence of claim 12.

15. An expression vector comprising the nucleotide sequence of claim 12.
