- a) a first plate, the first plate comprising;
a base layer of the first plate; and
one or more wells, wherein each well of the one or more wells comprises;
a side wall that extends vertically from the base layer;
an interior region of the well; and
an exterior region of the well, wherein the exterior region of the well has a lower surface energy than the interior region of the well; and
b) a second plate, wherein the second plate comprises a plurality of clusters, wherein each cluster comprises a plurality of loci, wherein each locus comprises a physically distinct region comprising molecules capable of binding to a nucleoside phosphoramidite, and wherein each cluster is vertically aligned to a single well of the one or more wells.

Abstract
De novo synthesized large libraries of nucleic acids are provided herein with low error rates. Further, devices for the manufacturing of high-quality building blocks, such as oligonucleotides, are described herein. Longer nucleic acids can be synthesized in parallel using microfluidic assemblies. Further, methods herein allow for the fast construction of large libraries of long, high-quality genes. Devices for the manufacturing of large libraries of long and high-quality nucleic acids are further described herein.

1. A device for nucleic acid assembly, comprising:

a) a first plate, the first plate comprising; a base layer of the first plate; and one or more wells, wherein each well of the one or more wells comprises; a side wall that extends vertically from the base layer; an interior region of the well; and an exterior region of the well, wherein the exterior region of the well has a lower surface energy than the interior region of the well; and b) a second plate, wherein the second plate comprises a plurality of clusters, wherein each cluster comprises a plurality of loci, wherein each locus comprises a physically distinct region comprising molecules capable of binding to a nucleoside phosphoramidite, and wherein each cluster is vertically aligned to a single well of the one or more wells. - View Dependent Claims (2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, 17)
