Abstract

The invention provides non-human cells and mammals having a genome encoding chimeric antibodies and methods of producing transgenic cells and mammals. Certain aspects of the invention include chimeric antibodies, humanized antibodies, pharmaceutical compositions and kits. Certain aspects of the invention also relate to diagnostic and treatment methods using the antibodies of the invention.

Claims

1. A synthetic polynucleotide comprising a gene segment comprising an exon encoding an immunoglobulin light chain variable (V) polypeptide and an immunoglobulin light chain non-coding element, wherein the V polypeptide is from a first species, wherein the non-coding element is from a second species, and wherein the exon encoding the V polypeptide replaces a V coding sequence of the second species.

2. The synthetic polynucleotide of claim 1, wherein the first species is human.

3. The synthetic polynucleotide of claim 2, wherein the second species is a non-human mammal.

4. The synthetic polynucleotide of claim 2, wherein the second species is mouse.

5. The synthetic polynucleotide of claim 1, wherein the non-coding element is selected from an intron or a cis regulatory element.

6. The synthetic polynucleotide of claim 5, wherein the cis regulatory element is selected from a promoter, an enhancer, a recombination signal sequence and a 5′

untranslated region.

7. The synthetic polynucleotide of claim 1, further comprising a gene segment encoding an immunoglobulin light chain joining (J) polypeptide from the first species, wherein the gene segment encoding the J polypeptide replaces an immunoglobulin light chain J coding sequence of the second species.

8. The synthetic polynucleotide of claim 1, wherein the V exon encodes a kappa light chain V (Vκ

) polypeptide or a lambda light chain V (Vλ

) polypeptide.

9. The synthetic polynucleotide of claim 8, wherein the V exon is in germline configuration.

10. The synthetic polynucleotide of claim 8, wherein the gene segment comprising the V exon is capable of undergoing rearrangement to produce a polynucleotide sequence encoding a polypeptide comprising an immunoglobulin light chain variable region.

11. The synthetic polynucleotide of claim 1, wherein the transgene is a synthetic transgene.

12. The synthetic polynucleotide of claim 11, wherein the non-coding element and the V polypeptide are from non-orthologous immunoglobulin loci.

13. The synthetic polynucleotide of claim 12, wherein the non-coding element is from an immunoglobulin heavy chain locus.

The synthetic polynucleotide of claim 1, wherein the transgene further comprises a gene segment encoding an immunoglobulin light chain constant (C) polypeptide, wherein the C polypeptide is from the first species, and wherein the gene segment encoding the C polypeptide replaces an immunoglobulin light chain C coding sequence of the second species.

15. The synthetic polynucleotide of claim 14, wherein the C gene segment encodes a Cλ

polypeptide or a Cκ

polypeptide.

16. The synthetic polynucleotide of claim 1, wherein an immunoglobulin light chain 3′

enhancer from the second species is replaced with an immunoglobulin light chain 3′

enhancer from the first species.

17. A mammalian cell whose genome comprises a transgene comprising the synthetic polynucleotide of claim 1.

18. A non-human mammal whose genome comprises a transgene comprising the synthetic polynucleotide of claim 1.
