-disulfophenylimino)-3H-phenothiazine mediator, the method comprising;dissolving phenothiazine in tetrahydrofuran to form a phenothiazine solution; dissolving analine 2,5-disulfonic acid in water and sodium hydroxide to form an analine 2,5-disulfonic acid solution; mixing the analine 2,5-disulfonic acid solution with the phenothiazine solution to form a phenothiazine/analine suspension; mixing the phenothiazine/analine suspension with a sodium persulfate solution to form a mixture; evaporating the tetrahydrofuran from the mixture to form a remaining solution; backwashing the remaining solution with ethyl acetate to form a reaction fluid; cooling the reaction fluid to precipitate solids therefrom; and mixing the precipitated solids with acetonitrile to form the 3-(2′
