Abstract

Disclosed is a pharmaceutical composition for treating hypercholesterolemia. The pharmaceutical composition includes a polysaccharide-polyamine copolymer and a pharmaceutically acceptable salt thereof as active ingredients. The polysaccharide-polyamine copolymer is formed by copolymerization of the following two parts: a selectively oxidized polysaccharide with 2,3-dialdehyde, and a polyamine with an amino functional group; the polyamine with an amino functional group and the selectively oxidized polysaccharide with 2,3-dialdehyde can form a net structure by means of covalent crosslinking, resulting in a hydrogel with an amino functional group or a granular polysaccharide-polyamine copolymer, wherein the amino functional group in the hydrogel with an amino functional group or the granular polysaccharide-polyamine copolymer can be protonated so as to form a cationic copolymer of a three-dimensional network structure having a protonated site, and the nitrogen content of the cationic copolymer and the nitrogen content of the polysaccharide-polyamine copolymer are above 12.3 wt %, and both the cationic copolymer and the polysaccharide-polyamine copolymer are water-insoluble.

40. A pharmaceutical composition for treatments of hypercholesterolemia, the pharmaceutical composition comprises an amino-polysaccharide copolymer and a pharmaceutically acceptable salt thereof as an active ingredient, the amino-polysaccharide copolymer is a copolymerization product of the following two parts:

a selectively oxidized polysaccharide having a 2,3-dialdehyde moiety, and a amino polymer which provides an amino functionality;

the amino polymer cross linking the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety to form a net-like structure and finally to provide a hydrogel having an amino functionality or a particulate amino-polysaccharide copolymer, amino functionalities of the hydrogel having an amino functionality or the particulate amino-polysaccharide copolymer can be protonated to form a cationic copolymer having three dimensional network structure and protonation sites, a nitrogen content of the cationic copolymer or the amino-polysaccharide copolymer is at least 12.3% weight percent, or at least 15% weight percent, or at least 22% weight percent, or at least 40% weight percent, the cationic copolymer and the amino-polysaccharide copolymer are both insoluble in water.

41. The pharmaceutical composition according to claim 40, wherein a nitrogen content of the amino polymer which provides an amino functionality is at least 24.5% weight percent, or at least 30% weight percent, or at least 44% weight percent, or at least 80% weight percent.

42. The pharmaceutical composition according to claim 40, wherein a molecular weight of the amino polymer which provides an amino functionality is from about 15,000 to about 900,000.

43. The pharmaceutical composition according to claim 40, wherein the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety is one or more selected from the group consisting of selectively oxidized cellulose, selectively oxidized amylopectin, and selectively oxidized chitosan.

44. The pharmaceutical composition according to claim 40, wherein in the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety, a ratio of an amount of oxidized glucose units to the total amount of glucose units is at least 50%, or at least 70%, or at least 80%.

45. The pharmaceutical composition according to claim 40, wherein the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety has at least one of a β

-1,4-glycosidic bond or a β

-1,6-glycosidic bond.

46. The pharmaceutical composition according to claim 45, wherein the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety has a β

-1,4-glycosidic bond and does not have a β

-1,6-glycosidic bond.

47. The pharmaceutical composition according to claim 40, wherein the amino polymer which provides an amino functionality is one or more selected from the group consisting of polyethyleneimine (PEI), poly(allylamine), polypropyleneimine (PPI), and polypropylenimine tetramine.

48. The pharmaceutical composition according to claim 40, wherein the amino polymer which provides an amino functionality has a branched structure or a dendritic structure.

49. The pharmaceutical composition according to claim 40, wherein a size of the hydrogel having an amino functionality or the particulate amino-polysaccharide copolymer or the cationic copolymer is from 30 μ

m to 10 mm, or from 100 μ

m to 10 mm, or from 300 μ

m to 5 mm.

50. The pharmaceutical composition according to claim 40, wherein the hydrogel having an amino functionality or the particulate amino-polysaccharide copolymer or the cationic copolymer has pores with diameters less than 50 μ

m, or has pores with diameters from 100 nm to 50 μ

m, or has pores with diameters from 200 nm to 40 μ

m, or has pores with diameters from 300 nm to 30 μ

m, or has pores with diameters from 400 nm to 20 μ

m, or has pores with diameters from 500 nm to 10 μ

m, or has pores with diameters from 800 nm to 5 μ

m.

51. A use of a pharmaceutical composition comprising an amino-polysaccharide copolymer and a pharmaceutically acceptable salt thereof as an active ingredient in the preparation of a medicament for the treatment of hypercholesterolemia, wherein, the amino-polysaccharide copolymer is a copolymerization product of the following two parts:

a selectively oxidized polysaccharide having a 2,3-dialdehyde moiety, and a amino polymer which provides an amino functionality;

the amino polymer cross linking the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety to form a net-like structure and finally to provide a hydrogel having an amino functionality or a particulate amino-polysaccharide copolymer, amino functionalities of the hydrogel having an amino functionality or the particulate amino-polysaccharide copolymer can be protonated to form a cationic copolymer having three dimensional network structure and protonation sites, a nitrogen content of the cationic copolymer or the amino-polysaccharide copolymer is at least 12.3% weight percent, or at least 15% weight percent, or at least 22% weight percent, or at least 40% weight percent, the cationic copolymer and the amino-polysaccharide copolymer are both insoluble in water.

52. The use according to claim 51, wherein a nitrogen content of the amino polymer which provides an amino functionality is at least 24.5% weight percent, or at least 30% weight percent, or at least 44% weight percent, or at least 80% weight percent.

53. The use according to claim 51, wherein a molecular weight of the amino polymer which provides an amino functionality is from about 15,000 to about 900,000.

54. The use according to claim 51, wherein the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety is one or more selected from the group consisting of selectively oxidized cellulose, selectively oxidized amylopectin, and selectively oxidized chitosan.

55. The use according to claim 51, wherein in the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety, a ratio of an amount of oxidized glucose units to the total amount of glucose units is at least 50%, or at least 70%, or at least 80%.

56. The use according to claim 51, wherein the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety has at least one of a β

-1,4-glycosidic bond or a β

-1,6-glycosidic bond.

57. The use according to claim 56, wherein the selectively oxidized polysaccharide having a 2,3-dialdehyde moiety has a β

-1,4-glycosidic bond and does not have a β

-1,6-glycosidic bond.

58. The use according to claim 51, wherein the amino polymer which provides an amino functionality is one or more selected from the group consisting of polyethyleneimine (PEI), poly(allylamine), polypropyleneimine (PPI), and polypropylenimine tetramine.

59. The use according to claim 51, wherein the amino polymer which provides an amino functionality has a branched structure or a dendritic structure.

60. The use according to claim 51, wherein a size of the hydrogel having an amino functionality or the particulate amino-polysaccharide copolymer or the cationic copolymer is from 30 μ

m to 10 mm, or from 100 μ

m to 10 mm, or from 300 μ

m to 5 mm.

61. The use according to claim 51, wherein the hydrogel having an amino functionality or the particulate amino-polysaccharide copolymer or the cationic copolymer has pores with diameters less than 50 μ

m, or has pores with diameters from 100 nm to 50 μ

m, or has pores with diameters from 200 nm to 40 μ

m, or has pores with diameters from 300 nm to 30 μ

m, or has pores with diameters from 400 nm to 20 μ
