Abstract

The present invention relates to an EPRS (glutamyl-prolyl-tRNA synthetase) protein or a fragment thereof.

The EPRS protein of the present invention or fragment thereof may bind to PCBP2 protein to activate the MAVS signaling pathway, and thus it has anti-RNA viral effects, thereby being effective for preventing or treating a RNA viral infectious disease.

Claims

1. A method of preventing or treating a RNA viral infectious disease, comprising administering to a subject an EPRS (glutamyl-prolyl-tRNA synthetase) protein, a fragment thereof, or a composition of an EPRS protein or a fragment thereof, wherein the fragment of the EPRS protein comprises an amino acid sequence at positions 168 to 186 in SEQ ID NO:

2. The method of claim 1, wherein the fragment of the EPRS protein comprises an amino acid sequence at positions 168 to 186 in SEQ ID NO:

63 and consists of 19 to 682 amino acids.

The method of claim 1, wherein the fragment of the EPRS protein comprises an amino acid sequence selected from the group consisting of an amino acid sequence at positions 1 to 196, an amino acid sequence at positions 1 to 269, an amino acid sequence at positions 1 to 457, an amino acid sequence at positions 1 to 506, an amino acid sequence at positions 1 to 682, an amino acid sequence at positions 166 to 506, an amino acid sequence at positions 168 to 506, an amino acid sequence at positions 166 to 269, an amino acid sequence at positions 168 to 269, an amino acid sequence at positions 166 to 196, an amino acid sequence at positions 168 to 196, an amino acid sequence at positions 166 to 186, and an amino acid sequence at positions 168 to 186 in SEQ ID NO:

The method of claim 1, wherein the RNA virus is one or more selected from the group consisting of Amalgaviridae, Birnaviridae, Chrysoviridae, Cystoviridae, Endornoviridae, Hypoviridae, Megobirnaviridae, Partitiviridae, Picobirnoviridae, Reoviridae, Totiviridae, Quadriviridae, Arteriviridae, Coronaviridae, Mesoniviridae, Roniviridae, Dicistroviridae, ifloviridae, Momoaviridae, Picornoviridae, Secoviridae, Alphaflexiviridae, Betoflexiviridae, Gammoflexiviridae, Tymoviridae, Bornaviridae, Filoviridae, Paramyxoviridae, Rhobdoviridae, Nyamiviridae, Coliciviridae, Flaviviridae, Luteoviridae, Togaviridae, Pneumoviridae, Arenoviridae, Deltavirus, and Orthomyxoviridae viruses.

5. The method of claim 1, wherein the EPRS protein or the fragment thereof is bound to a drug delivery vehicle.

6. The method of claim 5, wherein the drug delivery vehicle is one or more selected from the group consisting of a labeling material, a targeting ligand, a cell-penetrating peptide, a polymeric nanoparticle, a viral vector, a virus-like particle, and an inorganic nanoparticle.

7. The method of claim 6, wherein the cell-penetrating peptide is one or more selected from the group consisting of Tat, Antennapedia, Transportan, VP22, Hph-1, R11(R9), a signal sequence-based peptide, and an amphipathic peptide.

8. A method of preventing or inhibiting MAVS protein degradation comprising administering to a subject an EPRS (glutamyl-proplyl-tRNA synthetase) protein, a fragment thereof, or a composition comprising an EPRS protein or a fragment thereof, wherein the fragment of the EPRS protein comprises an amino acid sequence at positions 168 to 186 of SEQ ID NO:

12. The EPRS protein or fragment thereof of claim 9, wherein the EPRS protein or the fragment thereof is bound to a drug delivery vehicle.

13. The EPRS protein or fragment thereof of claim 12, wherein the drug delivery vehicle is bound to the EPRS protein or the fragment thereof via a linker.

The EPRS protein or fragment thereof of claim 12, wherein the drug delivery vehicle is one or more selected from the group consisting of a labeling material, a targeting ligand, a cell-penetrating peptide, a polymeric nanoparticle, a viral vector, a virus-like particle, and an inorganic nanoparticle.

15. The EPRS protein or fragment thereof of claim 14, wherein the cell-penetrating peptide is one or more selected from the group consisting of Tat, Antennapedia, Transportan, VP22, Hph-1, R11(R9), a signal sequence-based peptide, and an amphipathic peptide.

16. A polynucleotide encoding the EPRS protein or fragment thereof of claim 9.

17. A vector comprising the polynucleotide of claim 16.

18. A transformant comprising the polynucleotide of claim 16.

19. A composition comprising the EPRS protein of fragment thereof of claim 9.

20. The composition of claim 19, wherein the composition further comprises a non-naturally occurring carrier.
