1. A method for delivering from an osmotic device a drug that is difficult to deliver to an environment of use having a pH of less tha seven, wherein the method comprises:
- (1) admitting into the environment an osmotic drug delivery device, which device comprises;
(a) a shaped wall formed of a semipermeable material that is permeable to the passage of fluid present in the environment, and substantially impermeable to the passage of drugs and compounds, which wall surrounds and forms;
(b) a compartment housing a drug that is difficult to deliver to the environment, and a basic compound having a gas generating moiety, which compound is soluble in fluid that enters the compartment;
(c) a passageway in the wall connecting the compartment and the exterior of the device for dispensing drug from the device; and
,(2) imbibing fluid through the wall into the compartment in a tendency towards osmotic equilibrium at a rate determined by the permeability of the wall and the osmotic pressure gradient across the wall;
thereby(3) forming a solution of the basic compound containing drug, which solution is dispensed from the compartment through the passageway into the environment with the compound reactively;
(4) generating a gas on its contact with the fluid of the environment producing an effervescent suspension that delivers the drug in a finely dispersed form to the environment over a prolonged period of time.

Abstract
A method is disclosed for delivering a drug substantially-free of rapid precipitation from an osmotic device. The osmotic device comprises a semipermeable wall surrounding a compartment housing (1) a drug that exhibits limited solubility under neutral and acid conditions, and (2) a compound capable of evolving carbon dioxide in the presence of an acid in the environment of use. The method comprises, (a) imbibing fluid through the wall into the compartment at a rate determined by the permeability of the wall and the osmotic pressure gradient across the wall, (b) forming in the compartment a basic solution containing drug and compound, which solution is delivered from the compartment through the passageway, (c) reacting the compound with the acid in the environment at the device-environment interface for evolving carbon dioxide, thereby, (d) providing an effervescent suspension in the environment that delivers the drug in a finely dispersed form to the environment of use over time. Also, a composition is disclosed comprising the drug and the compound.

4. A method for dispersing a drug in an acid environment of use, wherein the method comprises:

(1) admitting into the acid environment an osmotic device comprising; (a) a shaped wall formed of a semipermeable polymeric composition that is permeable to the passage of fluid, and substantially impermeable to the passage of drug, with the wall surrounding and forming; (b) a compartment containing a drug that exhibits limited solubility in the acid environment, and a non-toxic basic compound having a carbon dioxide producing group, which compound is soluble in fluid that enters the compartment and exhibits an osmotic pressure gradient across the wall against an exterior fluid; (c) a passageway in the wall for dispensing drug from the device; and
,(2) dissolving the compound in fluid imbibed through the semipermeable wall into the compartment to produce a solution of the compound containing drug, which solution is dispensed through the passageway to the environment wherein the compound reacts with the acid environment and generates carbon dioxide gas that disperses the drug away from the device to the environment over time. - View Dependent Claims (5, 6, 7, 8, 9, 10)

11. A method for continuously administering an active drug having limited solubility in gastric fluid to the gastric environment of a warm blooded animal, comprising:
-
(1) admitting into the gastric environment an osmotic drug delivery device comprising; (a) a shaped wall formed of a semipermeable material that is permeable to the passage of an exterior fluid and substantially impermeable to the passage of drug, the wall surrounding and forming; (b) a compartment containing the drug, and a pharmaceutically acceptable compound selected from the group consisting of carbonates, bicarbonates and mixtures thereof, which compound is capable of effervescing in the presence of gastric fluid, is soluble in fluid that enters the compartment and exhibits an osmotic pressure gradient across the wall against the fluid; (c) a passageway in the wall for dispensing the drug from the device; and (2) imbibing fluid into the compartment through the wall at a rate determined by the permeability of the wall and the osmotic pressure gradient across the wall to form a solution containing drug that is dispensed at a controlled and continuous rate through the passageway to the gastric environment, wherein the carbonate reactively effervesces with the gastric fluid;

24. A method for substantially preventing the precipitation of a drug in the passageway of an osmotic device present in an acidic environment, which method comprises
(1) imbibing fluid present in the environment into the osmotic device, which device comprises: -
(a) a shape retaining wall formed of a semipermeable polymer selected from the group consisting of cellulose acetate, cellulose diacetate and cellulose triacetate that is permeable to the passage of an exterior fluid, and substantially impermeable to the passage of drugs and inorganic compounds, said wall surrounding and defining; (b) a compartment housing a drug that precipitates in a neutral or acid environment, and an inorganic compound having a gas generating moiety, which compound is soluble in fluid that enters the compartment; (c) a passageway in the semipermeable wall connecting the compartment with the exterior of the device for releasing the drug from the compartment; (2) forming in the compartment a solution of the compound containing the drug, which solution is dispensed from the compartment through the passageway into the environment; and (3) reacting the compound on its contact with the fluid of the environment, thereby producing a gas that substantially prevents the precipitation of drug in the passageway of the device.

25. A method for substantially preventing, in the environment of use, precipitation of drug on the exterior surface of an osmotic device, which method comprises:
-
(1) admitting into the environment an osmotic device, which device comprises; (a) a shaped wall formed of a semipermeable polymer that is permeable to the passage of an exterior fluid, and substantially impermeable to the passage of drugs and compounds, said wall surrounding and forming; (b) a compartment; (c) a passageway in the wall connecting the compartment with the exterior of the device; and (d) a drug in the compartment that precipitates in the environment on the exterior surface of the device in the vicinity of the passageway, and a basic compound having a gas generating moiety in the compartment, which compound is soluble in fluid that enters the compartment; (2) imbibing fluid through the wall into the compartment from the environment in a tendency towards osmotic equilibrium at a rate determined by the permeability of the wall and the osmotic pressure gradient across the wall;
thereby,(3) forming a solution of compound containing drug, which solution is dispensed from the compartment through the passageway into the environment; and
,(4) producing a gas by reacting the compound on its contact with the environment thereby raising the pH in the environment in the vicinity of the passageway, which gas and pH substantially prevent the precipitation of drug on the exterior of the device. - View Dependent Claims (26, 27)

28. A method for continuously administering a pharmaceutically acceptable drug to an acidic body fluid of an animal, which method comprises:
-
(1) admitting into the body fluid an osmotic delivery device sized, shaped and adapted for easy admittance and prolonged placement in the body fluid, said device comprising; (a) a wall formed of a semipermeable polymer that is permeable to the passage of the body fluid and substantially impermeable to the passage of body fluid, which wall surrounds and defines; (b) a compartment containing the drug, and a non-toxic compound selected from the group consisting of a carbonate, bicarbonate, and mixtures thereof, which compound produces effervescence in the presence of acid fluid, is soluble in fluid that enters the compartment and exhibits an osmotic pressure gradient across the semipermeable wall against the fluid; (c) a passageway in the wall for delivering the drug from the compartment; and
,(2) imbibing fluid into the compartment through the wall at a rate determined by the permeability of the wall and the osmotic pressure gradient across the wall to form a solution containing drug that is delivered at a controlled and continuous rate through the passageway to the acidic body fluid, wherein the compound reacts with the acidic fluid and effervescence;
thereby,(3) delivering the drug in a therapeutically effective amount to the animal over a prolonged period of time. - View Dependent Claims (29)

30. A method for treating inflammation in a warm-blooded animal, which method comprises:
-
(1) admitting orally into the gastric environment of the animal an osmotic delivery device comprising; (a) a wall formed of a non-toxic semipermeable polymer that is permeable to the passage of gastric fluid and substantially impermeable to the passage of an antiinflammatory drug, which wall surrounds and forms; (b) a compartment housing a dosage unit amount of an antiinflammatory drug that precipitates in the gastric environment, and a non-toxic basic compound having a carbon dioxide generating moiety; (2) imbibing fluid through the semi-permeable wall into the compartment in a tendency towards osmotic equilibrium, at a rate determined by the permeability of the wall and the osmotic pressure gradient across the wall, to form a solution of basic compound containing drug, that is dispensed through the passageway into the gastric environment; and
,(3) generating carbon dioxide by reacting the compound with fluid in the gastric environment, said gas substantially preventing the precipitation of the antiinflammatory drug in the vicinity of the device, thereby delivering a therapeutically effective amount of the antiinflammatory drug for treating the inflammation over a prolonged period of time. - View Dependent Claims (31)
