Abstract

Apparatus and method for forming sealed pharmaceutical capsules containing precisely compacted charges of a powdered material. A pair of substantially cylindrical sealing rolls are arranged in nipping relationship for directing a pair of webs of gelatin material along predetermined paths between the nip of the sealing rolls. For each sealing roll there is provided a powder supply hopper which contains a supply of powdered material and a cylindrical roll rotatably mounted adjacent the supply hopper. The cylindrical roll has chambers extending radially inwardly from the surface thereof for receiving therewith powdered material as the cylindrical roll rotates past the supply hopper. A compaction mechanism is associated with each cylindrical roll for compacting the predetermined charge of powdered material in the chambers so as to form a compacted slug. A transfer roll associated with each cylindrical roll is provided intermediate the cylindrical roll and its respective sealing roll for transferring the compacted slugs of material to cavities formed in the web as the sealing roll feeds the web along its predetermined path toward the nip between the pair of sealing rolls, the transfer rolls being provided with means for collecting free and uncompacted powdered material and dust to minimize the depositing of dust and free powdered material onto the web.

Claims

1. Apparatus for forming a pharmaceutical capsule containing a powdered material therein, said apparatus comprising:

feeding means for feeding a capsule forming material having a cavity formed therein along a predetermined path, said capsule forming material forming a portion of the casing of the capsule;

powder supply means containing a supply of powdered pharmaceutical material;

a cylindrical punch roll having a radially extending chamber therein extending radially inward from an open end on the cylindrical surface of said cylindrical punch roll, said cylindrical punch roll being rotatably mounted adjacent said supply means to receive within said chamber a predetermined charge of powdered material from said supply means as said cylindrical punch roll rotates past said supply means, and said powder supply means and said cylindrical punch roll both being operatively isolated from said predetermined path along which said capsule forming material is fed;

punch compacting means associated with said cylindrical punch roll, said punch compacting means including punch means for compacting said predetermined charge of powdered material in said chamber to form a compacted substantially solid slug as said cylindrical punch roll is rotated;

a cylindrical transfer roll associated with said cylindrical punch roll for transferring said compacted substantially solid slug in said chamber of said punch roll into said cavity in said capsule forming material as said capsule forming material is fed along said predetermined path, said transfer roll having a recess in the cylindrical surface thereof and being rotatably mounted adjacent to said cylindrical punch roll and intermediate said cylindrical punch roll and said predetermined path along which said capsule forming material is fed, said transfer roll being operatively rotatable to align said recess therein with said chamber in said punch roll as said punch roll is rotated after said compacted substantially solid slug is formed in said chamber to receive said compacted substantially solid slug from said chamber into said recess and operatively rotatable to then deposit said compacted substantially solid slug in said recess into said cavity in said capsule forming material;

said transfer roll having means associated therewith for collecting free and uncompacted powdered material transferred with said compacted substantially solid slug to said transfer roll to thereby minimize the depositing of free, excess powdered material onto said capsule forming material; and

sealing means for sealing said cavity in said capsule forming material after it has received said compacted substantially solid slug.

The apparatus of claim 1 wherein said means associated with said transfer roll for collecting free and uncompacted powdered material comprises a vacuum means communicating with said recess in said cylindrical surface of said transfer roll for applying a reduced pressure to said recess during a portion of the rotation of said transfer roll.

The apparatus of claim 2 wherein said vacuum means communicates with said recess in a manner such that said compacted substantially solid slug transferred into said recess is maintained in said recess during said portion of the rotation of said transfer roll during which said reduced air pressure is applied to said recess.

4. The apparatus of claim 3 wherein said transfer roll comprises a stationary core member, a rotatable annular sleeve member having said recess in the outer cylindrical surface thereof and having an inner cylindrical surface surrounding said stationary core member, means defining a circumferential cavity between said rotatable annular sleeve member and said stationary core member about a portion of the circumference of said stationary core member, and a passageway extending between said recess and said inner cylindrical surface of said rotatable annular sleeve member to provide communication between said recess and said circumferential cavity during said portion of the rotation of said transfer roll;

and wherein said vacuum means comprises a source of vacuum communicating with said circumferential cavity.

5. The apparatus of claim 4 wherein said circumferential cavity comprises a first circumferential cavity about a first portion of the circumference of said stationary core member;

and further including means defining a second circumferential cavity between said rotatable sleeve member and said stationary core member about a second portion of the circumference of said stationary core member displaced from said first portion, said second circumferential cavity being disposed about said stationary core member so that when said recess in said rotatably sleeve member is aligned with said cavity in said capsule forming material, said passageway is in communication with said second circumferential cavity, and a source of pressurized gas communicating with said second circumferential cavity.

The apparatus of claim 1 wherein said punch means of said compacting means comprises a movable punch member disposed in said chamber of said punch roll for movement toward and away from the surface of said punch roll, and punch member moving means for moving said punch member toward and away from said cylindrical surface of said punch roll as said punch roll is rotated;

and wherein said compacting means further includes closure means disposed in operative relationship to said punch roll to close said open end of said chamber after said chamber has received said predetermined charge of powdered material; and

wherein said punch member moving means is operative to move said punch member toward said cylindrical surface of said punch roll after said closure means has closed said open end of said chamber to compact said powdered material against said closure means.

7. The apparatus of claim 6 wherein said supply means is supported at an elevation above said punch roll and supplies powdered material to the surface of said punch roll so that as said punch roll rotates, powdered material falls into said chamber in said punch roll;

and wherein said punch member moving means is operative to move said punch member to a predetermined position within said chamber as said punch roll rotates past said supply means so that the volume in said chamber between said punch member and the surface of said punch roll corresponds to the volume of said predetermined charge of powdered material.

8. The apparatus of claim 7 further including first adjustment means for adjusting the position of said punch member in said chamber as said punch roll rotates past said supply means to change said predetermined position of said punch member in said chamber to thereby adjust said predetermined charge of powdered material received within said chamber.

The apparatus of claim 8 further including second adjustment means for adjusting the extent of movement of said punch member toward said open end of said chamber when said closure means is operative to close said open end of said chamber to thereby adjust the degree of compaction of said powdered material.

10. The apparatus of claim 9 wherein said punch member includes a cam head thereon, and wherein said first adjustment means comprises a first nonrotating cam member supported relative to the axis of rotation of said punch roll, and means for adjusting the position of said first cam member relative to said axis of rotation of said punch roll, said first cam member being operatively disposed to be engaged by said cam head as said open end of said chamber in said punch roll moves past said supply means.

11. The apparatus of claim 10 wherein said second adjusting means comprises a second nonrotating cam member supported relative to said axis of rotation of said punch roll, and means for adjusting the position of said second cam member relative to said axis of said punch roll, said second cam member being operatively positioned to be engaged by said cam head on said punch member when said open end of said chamber is closed by said closure means.

12. The apparatus of claim 11 wherein said punch member moving means comprises an annular cam track for said cam head of said punch member, said cam track being stationarily supported relative to the axis of rotation of said punch roll and having an open portion at the circumferential location of said first and second cam members so that said cam head is engagable by said first and second cam members as it moves therepast as said punch roll is rotated.

13. The apparatus of claim 12 wherein said first and second nonrotating cam members and said cam track are stationarily positioned inside said cylindrical surface of said cylindrical roll.

14. The apparatus of claim 13 wherein said cylindrical punch roll includes a stationary core member and a rotatable sleeve member rotatably positioned thereon, said rotatable sleeve member having said radially extending chamber therein, and wherein said first and second nonrotating cam members and said cam track are supported by said stationary core member.

The apparatus of claim 14 wherein said first and second nonrotating cam members each have an axially extending bore therethrough extending in a direction parallel to the axis of rotation of said punch roll, wherein said means for adjusting the position of said first cam member comprises a first eccentrically mounted rotatable disc member positioned in said axially extending bore of said first nonrotating cam member and rotatable to thereby change the radial position of said first cam member, and wherein said means for adjusting the position of said second cam member relative to the axis of said punch roll comprises a second eccentrically mounted rotatable disc member positioned in said axially extending bore of said second nonrotating cam member and rotatable to thereby change the radial position of said second cam member.

16. The apparatus of claim 15 wherein the radial dimension of said rotatable disc members is less than the respective inner dimensions of said axially extending bores, and further including means for each of said first and second cam members for urging a portion of the inner surface of said bores into engagement with the outer surface of its respective rotatable disc member.

17. The apparatus of claim 16 further including means for rotating said disc members from a position axially remote from said punch members.

18. The apparatus of claim 11 wherein said closure means comprises a surface closing member supported relative to the axis of rotation of said punch roll for engaging the surface of said punch roll as said punch roll rotates so that said open end of said chamber in said surface of said punch roll is closed by said surface closing member as said open end moves therepast.

19. The apparatus of claim 18 wherein said surface closing member comprises a belt member trained about first and second drive rollers and operatively disposed with respect to a portion of the circumferential surface of said punch roll along the axial length thereof and in engagement therewith.

20. The apparatus of claim 6, further including adjustment means for adjusting the position of said punch member in said chamber as said cylindrical punch roll rotates past said supply means so that the amount of material received within said chamber corresponds to said predetermined charge of material, said adjustment means being adjustable to vary the predetermined charge of material received within said chamber.

The apparatus of claim 20 wherein said adjustment means is operative to move said punch member to a first position which defines a volume in said chamber between said punch member and the surface of said cylindrical roll which is greater than the volume corresponding to said predetermined charge as said open end of said chamber is rotated past said supply means, and is then operative to move said punch member to a second position which defines a volume in said chamber between said punch member and the surface of said cylindrical roll which is substantially the same as said volume corresponding to said predetermined charge after said chamber has received powdered material.

22. The apparatus of claim 21 further including scraping means for scraping the surface of said punch roll to remove excess powder expelled from said chamber as said punch member moves from said first position to said second position.

23. The apparatus of claim 22 further including punch member moving means for moving said punch member along a predetermined punch path as said punch roll is rotated;

and wherein said adjustment means is operative to vary said predetermined punch path as said chamber is moved past said powder supply means.

24. The apparatus of claim 6 wherein said punch roll includes a plurality of radially extending chambers therein extending radially inward from the cylindrical surface of said punch roll, said chambers being arranged circumferentially about the surface of said punch roll and axially along the length of said punch roll;

wherein said punch means of said compacting means comprises a plurality of movable punch members disposed in said chambers for movement towards and away from the surface of said cylindrical roll; and

wherein said punch member moving means is operative to move said plurality of punch members towards and away from the surface of said punch roll.

25. The apparatus of claim 1 wherein said feeding means comprises a rotatable sealing roll about which a web of capsule forming material is trained, said sealing roll having a plurality of recesses therein to form a plurality of cavities in said web as said sealing roll is rotated;

and wherein said cylindrical punch roll, said compacting means and said transfer roll are operative to transfer compacted substantially solid slugs of powdered material into said plurality of cavities in said web.

26. The apparatus of claim 25 wherein said cylindrical punch roll includes a plurality of said chambers on the cylindrical surface thereof for receiving predetermined charges of powdered material therein from said supply means as said cylindrical punch roll rotates past said supply means;

wherein said compacting means is operative to compact said predetermined charges of powdered material in each of said chambers to form compacted substantially solid slugs as said cylindrical punch roll rotates; and

wherein said transfer roll is operative to transfer said formed compacted substantially solid slugs into said cavities in said web as said web is moved along said predetermined path by said sealing roll.

27. The apparatus of claim 26 wherein said sealing roll comprises a first sealing roll and said web comprises a first web;

wherein said sealing means comprises a second sealing roll arranged in nipping relationship to said first sealing roll and having a second web of capsule forming material trained thereabout, said second sealing roll being rotatable to move said second web into overlying relationship with respect to said first web of capsule forming material and to seal said webs together as said webs pass between the nip of said sealing rolls.

28. The apparatus of claim 27 wherein said second sealing roll has a plurality of recesses therein for forming cavities in said second web as said second sealing roll is rotated;

and wherein first and second sealing rolls are arranged and rotated so that said cavities in said first web are brought into juxtaposition with said cavities in said second web at the nip between said first and second sealing rolls and sealed together around said cavities, the surface of said cavities forming the inner surfaces of said sealed capsules.

29. The apparatus of claim 28 further including depositing means for depositing a predetermined charge of powdered pharmaceutical material into said cavities in said second web as said second sealing roll rotates.

30. The apparatus of claim 29 wherein said powder supply means comprises first powder supply means;

said cylindrical punch roll comprises a first cylindrical punch roll;

said compacting means comprises a first compacting means including first punch means; and

said transfer roll comprises a first transfer roll; and

wherein said depositing means for depositing material into said second web comprises second powder supply means containing a supply of powdered pharmaceutical material;

a second cylindrical punch roll having a plurality of radially extending chambers therein extending radially inward from an open end on the cylindrical surface of said second cylindrical punch roll, said second cylindrical punch roll being rotatably mounted adjacent said second supply means to receive within said chamber said predetermined charges of powdered material from said second powder supply means as said second cylindrical punch roll rotates past said second powder supply means;

second punch compacting means associated with said second cylindrical punch roll, said second punch compacting means including second punch means for compacting said predetermined charges of powdered material in said chambers of said second cylindrical punch roll to form compacted substantially solid slugs therein; and

a second transfer roll associated with said second cylindrical punch roll for transferring said compacted substantially solid slugs in said chambers of said second cylindrical punch roll into said cavities in said second web as said second web is fed by said second sealing roll.

31. A method of forming a pharmaceutical capsule containing a powdered material therein, said method comprising the steps of:

feeding a capsule forming material having a cavity formed therein along a predetermined path, the capsule forming material forming a portion of the casing of the capsule;

rotating a cylindrical punch roll having a radially extending chamber therein extending radially inward from an open end on the cylindrical surface of said cylindrical punch roll, said cylindrical punch roll being operatively isolated from said predetermined path along which said capsule forming materal is fed;

supplying a predetermined charge of powdered pharmaceutical material into said chamber as said cylindrical punch roll is rotated;

compacting said predetermined charge of powdered material in said chamber with punch means to form a compacted substantially solid slug as said cylindrical punch roll is rotated;

providing a rotatable cylindrical transfer roll intermediate said cylindrical punch roll and said predetermined path along which said capsule forming material is fed, said transfer roll including a recess in the cylindrical surface thereof;

operating said transfer roll to rotate in timed relationship to rotation of said cylindrical punch roll so as to align said recess in said transfer roll with said chamber in said cylindrical punch roll as said recess and said chamber pass through the nip between said punch roll and said transfer roll;

transferring said compacted substantially solid slug from said chamber into said recess in said transfer roll when said recess and said chamber are aligned;

rotating said transfer roll after said compacted substantially solid slug has been transferred into said recess to align said recess with said cavity in said capsule forming material;

depositing said compacted substantially solid slug into said cavity in said capsule forming material when said recess is aligned with said cavity;

collecting free and uncompacted powdered material transferred with said compacted substantially solid slug to said transfer roll during a portion of said rotating step to thereby minimize the depositing of free, excess powdered material onto said capsule forming material during said depositing step; and

sealing said cavity in said capsule forming material after it has received said compacted substantially solid slug.

32. The method of claim 31, wherein said collecting step comprises applying a reduced pressure to said recess in said transfer roll during said portion of said rotating step.

33. The method of claim 32 wherein said step of applying comprises applying a reduced pressure to said recess in a manner such that said compacted substantially solid slug transferred into said recess is maintained in said recess during said portion of said rotating step.

34. The method of claim 33 in which said transfer roll comprises a stationary core member, a rotatable annular sleeve member having said recess in the outer cylindrical surface thereof and having an inner cylindrical surface surrounding said stationary core member, means defining a circumferential cavity between said stationary core member and said rotatable sleeve member about a portion of the circumference of said stationary core member, and a passageway in said rotatable sleeve member extending between said recess and said inner cylindrical surface to provide communication between said circumferential cavity and said recess during said portion of said rotating step, and wherein said step of applying a reduced pressure to said recess comprises providing a source of vacuum communicating with said circumferential cavity.

35. The method of claim 34 in which said punch means comprises a movable punch member disposed in such chamber of said punch roll for movement towards and away from the surface of said cylindrical punch roll;

and wherein said step of compacting comprises closing said open end on the surface of said cylindrical punch roll as said cylindrical punch roll is rotated, and then moving said punch member toward the surface of said cylindrical punch roll to compact said predetermined charge of powdered material.

36. The method of claim 35 wherein said step of supplying a predetermined charge of powder into said chamber comprises supporting a powder supply means at an elevation above said cylindrical punch roll and supplying powdered material to the surface of said cylindrical punch roll so that powdered material falls into said chamber as said chamber moves past said powder supply means, and then moving said punch member to a predetermined position within said chamber as said cylindrical roll rotates past said supply means so that the volume in said chamber between said punch member and the cylindrical surface of said cylindrical punch roll corresponds to the volume of said predetermined charge of powdered material.

37. The method of claim 36 further including the step of first moving said punch member to a retracted position prior to said step of moving said punch member to said predetermined position as said chamber moves past said powder supply means, said punch member when in said retracted position being further from the cylindrical surface of said cylindrical roll than when said punch member is in said predetermined position.

38. The method of claim 37 wherein said steps of moving said punch member comprise mounting a stationary cam track within said cylindrical roll about which said cylindrical punch roll rotates, and providing a cam head on said punch member for receipt within said cam track so that said cam head follows said cam track to adjust the position of said punch member as said cylindrical punch roll rotates relative to said cam track.

39. The method of claim 31 in which said capsule forming material has a plurality of cavities formed therein and in which said cylindrical punch roll has a plurality of chambers formed therein for receiving predetermined charges of powdered material;

and wherein said step of compacting comprises compacting said predetermined charges of powdered material in said chambers; and

wherein said step of transferring comprises transferring said compacted substantially solid slugs into said cavities in said capsule forming material.

40. The method of claim 39 wherein said step of feeding a capsule forming material along a predetermined path comprises passing a undeformed sheet of capsule forming material about a rotatable sealing roll having recesses formed therein, and deforming overlying portions of said sheet into said recesses to form said plurality of cavities as said sheet engages said sealing roll and is moved thereby.

41. The method of claim 40 in which said web comprises a first web;

and wherein said step of sealing comprises feeding a second web into juxtaposition over said first web after said cavities in said first web have received said compacted slugs.

42. The method of claim 41 in which said second web has a plurality of cavities formed therein;

and further including the steps of compacting predetermined charges of powder material to form compacted substantially solid slugs for said second web and depositing said compacted substantially solid slugs into said cavities of said second web; and

wherein said step of sealing comprises feeding said second web with said cavities formed therein having compacted substantially solid slugs deposited therein together with said first web so that said cavities in said first and second webs are arranged in juxtaposition to one another and sealed around said cavities to form said capsules.
